Juglans mandshurica Mamix. is one of precious northeast three hard broad-leaf tree species, it is the third century remnant species. Most of them are wilding. In recent years, it has large cultivation with its own function researching. With returning farmland to forest policy, the growth and the yield areas of J.mandshurica Mamix.tree increased. Therefore, it is necessary to study on the J .mandshurica Mamix .processing technology.The compose of fatty acids in the kernel oil of J.mandshurica Mamix .was analysed by gas chromatography. The oil consisted mainly of linoleic acid 60.11%, linolenic acid 11.51%, oleic acid 23.46%, stearic acid 1.14%, palmitinic acid 3.41%, peanut olefine acid 0.47%. The total unsaturated fatty acid was 96%.The results showed that the oil content of the kernel was 66.12%.Researched three extraction solvents, impregnant extraction, supercritical CO2 extraction, aqueous enzyme. Single factor and orthogonal test design were used to optimize the extracting conditions in the experimentation of impregnant extraction, the optimum extraction conditions were that the extractive temperature was 60°C, the extractive time was 2h, and the ratio of materiel to liquid was 1:5, under these conditions, the extractive ratio of oil was 56.53%. Single factor and orthogonal test design were used to optimize the extracting conditions in supercritical CO2 extraction, the optimum extraction conditions were that extractive pressure 35Mpa, extractive temperature 40°C, extractive time 4h, under these conditions, the extractive ratio of oil was 56.34%; Single factor and Response Surface Methodology (RSM) were used to optimize the extracting conditions in the experimentation of aqueous enzymatic method extraction, the optimum extraction conditions were that enzyme reaction time was 4.66, the ratio of materiel to liquid wasl:4.17, enzyme added quality was 2.58%, under these conditions, the extractive ratio of oil was 57.21%. Among the there extractive methods, the experimentation of aqueous enzymatic method extraction was the best, which is lowest cost and polluter.Investigate the oxidative stability of J .mandshurica Mamix .oil, the experimental result indicated that, with the ambient temperature elevating, the J.mandshurica Mamix.oil's acid value, the peroxide number, the carbonyl price adding remarkably, explained that the rancidity regulation became graveness; Metal ion; Cu2+ and Fe3+ accelerated J .mandshurica Mamix.oil's rancidity process, moreover , Cu2+ is badder than Fe3+. With comparing the protection form light condition to the direct light, the direct light could cause the fatty acid rapid rancidity. The anti-oxidized experiment detected that, increase oxidation inhibitor PG and VE, could validly guard against the rancidity which occurs under the higher temperature condition. According to the above findings, the J.mandshurica Mamix.oil should be saved in the low temperature, evades the light, simultaneously must avoid with metallic ion contact, such as the copper iron. In order to prevent fat to rancidity, lengthens the preservation time, could add right amount oxidation inhibitor PG, VE.Fed rats the oil of supercritical CO2 extraction, the serum cholesterol, triglyceride, high-density lipoprotein cholesterol and low-density lipoprotein cholesterol levels of the rats were measured. The results showed that, the oil can lower cholesterol, triglycerides and low-density lipoprotein cholesterol levels, increase high-density lipoprotein cholesterol levels, and reducing blood lipid was proved.
