Flavonoids is one of the secondary metabolites by plants and is a natural non-toxic material which has wide range of biological activity. Some flavonoids which have activity of antioxidant and hypolipidemic have broad application prospects to prevent atherosclerosis. In this study, Lycium ruthenicum Murr.in Xinjiang was selected and extracted the total flavonoids from its leaves. Anti-oxidation and hypolipidemic of the total flavonoids from Lycium ruthenicum Murr.were studied for the future medicine of preventing atherosclerosis and providing a theoretical basis for its development and utilization of Lycium ruthenicum Murr.. Main content and the results are as follows:1.The total flavonoids from the leaves of Lycium ruthenicum Murr. was identified by the color reaction and UV spectra. Determined contents of total flavonoids with spectrophotometry, it was found the best extraction conditions from the leaves of Lycium ruthenicum Murr.through single-factor test and multi-factor orthogonal test was 60% ethanol at 60℃for 40 minutes with the solution and solid ratio was 1:25, extraction ratio was 0.23% and contents of total flavonoids of crude production was 0.14%.2.The results showed that AB-8 macroporous resin is the most appropriate to purificate the total flavonoids from Lycium ruthenicum Murr through comparison of static adsorption ratio and desorption ratio of six kinds of resin. AB-8 macroporous resin was selected to separate and purificate the total flavonoids from Lycium ruthenicum Murr.,the best purification condition was the concentration of sample solution was 0.20mg/mL,it's velocity was 2.0mL/min,pH value was 5.0, and eluted with 95% ethanol from AB-8 resin,velocity of ethanol was 4.0mL/min,it's volume was 5 times of resin. By AB-8 resin purification,the recovery ratio was 79.82% and contents of the total flavonoids was 2.80% which is 20 times of the crude production, the results showed that the process was practical and could be extended.3.It was found that the total flavonoids from Lycium ruthenicum Murr. has the ability of reduction and elimination hydroxyl radical and anti-lipid peroxidation reaction through the chemical reaction system in vitro. The total flavonoids from Lycium ruthenicum Murr. could inhibited RBC hemolysis, enhanced the ability of mouse serum scavenging hydroxyl radical, inhibited MDA production in the liver of mice through biological macromolecules response system. 4.was tested in vivo: The total flavonoids of Lycium ruthenicum Murr.could reduced hyperlipidemia mice plasmaTC, TG and LDL-C levels,increased HDL-C levels, through in vivo experiment which showed that it has biological effection of hyperlipidemia. And also it was found that the total flavonoids from Lycium ruthenicum Murr. can improve the activity of the mouse plasma T-AOC, liver GSH-Px and SOD and reduce the contents of liver MDA,which showed that it has biological activity of antioxidantion and it is a new source to get drugs of preventing atherosclerosis.5.Rutin,quercetin,luteolin, isorhamnetin and kaempferol is the most common component which has anti-oxidation and hypolipidemic ability according literatures have reported.The total flavonoids of Lycium ruthenicum Murr. also contains these five kinds of components by thin layer chromatogram (TLC) and high-performance liquid chromatography(HPLC) analysis, the contents account for 0.8870%, 0.0414%, 0.0411%, 0.0258% and 0.0448% respectively,all the five kinds of flavonoids components reach to 1.04% in fine products and 37.14% of the total flavonoids from Lycium ruthenicum Murr.. Hypolipidemic and antioxidant activity of the total flavonoids from Lycium ruthenicum Murr. may be the five kinds of flavonoids which play a role.
