Transfer factor (TransferFactor,TF) as an immune modulator has been used for many years in clinic. And the existing formulations of TF is water injection and oral. The mechanism of TF is that TF transfer antigen-specific information and stimulate the immune system. Although the mechanism of TF is imprecise, the effect is significant in the clinical treatment of viral diseases , such as herpes zoster, recurrent herpes simplex, hepatitis, immune deficiency disorders, etc..The effect of TF's oral solution is exact, but the drawback like valid for a short, easy to grow bacteria, high production costs, is also obvious. In order to solve above problems. We, in this study, change oral solid dosage form into effervescent tablets with unchanged effect and dosage.High concentrations of TF stock solution must be got before making effervescent tablets. For this, we explore the method which combines self-dissolving enzyme and freeze thaw to break cells . With the method ,the concentration of stock solution increases from 1mg/mL to 8mg/mL measured by UV absorption spectrum and HPLC. The time of production process is shorten by the 3 times, and a lot of manpower, power and time are saved.In this study, the composition obtained from TF stock solution was made up of five components. The main components, peak 2 and peak 3, account for 70% of the total composition . The molecular weight of each peak was determined, peak a molecular weight of 4500±200 of peak 1, 3200±100 of peak 2, 1600±100 of peak 3 and under 1000 of peaks 4, 5. These data suggest that the TF's major components are peptides containing nucleotides with the molecular weight under 5000.We studied the preparation of effervescent tablets with a transfer factor liquid method, first made of citric acid and transfer factor complex, then add accessories effervescent agent, by mixing, granulating, whole, pressed, testing, packaging foam obtained Teng film. Established quality standards and quality tablet inspection procedures have been tested and all targets were achieved quality standards.
