In recent years, the environment hormone's harm and the monitoring method received the people's widespread attention, some sensitive, high selective, simple, economic, practical analysis method research attached importance to us. This research uses YES(Yeast Estrogen Screen)to examine the estrogen equivalent in different water body in Jinan including wastewater (wastewater inflow ,anaerobic digesters, aeration tank, wastewater outflow 2,wastewater outflow 1A); hospital water(health centers for women and children, Provincial hospital of Traditional Chinese Medicine);environmental water sample (the Huang gang upstream , the Huang gang downstream, Yu Qing lake, Dong ping main channel, Muli village, Peng village).In order to prove the sensitivity and selectivity of this method, we then use HPLC-MS method to determine 5 representative estrogen content (estrogen (E1), estradiol (E2) ,oestriol (E3) ,ethynyl estradiol (EE2), diethyl stilboestrol (DES)) in these water bodies. Through the determination, we know the sewage treatment water outflow still remained the high density environment hormone, then using the male zebrafish to conduct the biology effect research, we use real time PCR method to research sewage and the estrogen(E1) to the grown-up male zebrafish gene expression change(vtg, esr).The experimental results are as follows:First we use YES(Yeast Estrogen Screen) to detect.In all water samples, only hospital samples (health centers for women and children, Provincial hospital of Traditional Chinese Medicine) can examine the estrogen activity, the environment water samples have not been examined the estrogen activities including the rivers, lakes and the pollution outlet upstream and downstream. The sewage treatment plant water samples including wastewater anaerobic digesters, aeration tank, wastewater outflow 2,wastewater outflow 1A have not been examined the estrogen activities also. But in the wastewater inflow, the reason should be the water sample's toxicity is too high to cause the yeast die.This kind of strong toxic water sample should link the alternative examination method to determine estrogen content.Then we use HPLC-MS to examine 9 sampling points together.In the sewage treatment plant samples,5 kinds of estrogen content are not changed on the whole,the estrogen (E1) content is high (10ng/L);in the hospital water samples,the estrogen content are relatively high (in particular estradiol(E2), estrogen (E1), diethyl stilboestrol(DES)),higher than other rivers and the sewage treatment plant water sample. In the Huang gang upstream and the Huang gang downstream,the water samples remain very high (estrogen content is 10ng/L).We use estrogen (E1) to delect the grown-up male zebrafish,using real time PCR to study vtg and the esr gene expression changes. Then we use the grown-up male zebrafish to study vtg and the esr gene expression change, dividing 3 density groups (wastewater inflow ,wastewater outflow 2,wastewater outflow 1A).All the wastewater samples are able to rise vtg and esr expression level,and are highest in 5 day,then drop.For the esr, wastewater inflow and wastewater outflow 1A are all highest expressed in the 5 day,but wastewater outflow 2 are highest in the 10 day.
