Objective: The aim of this research is the biosynthesis allicin through alliin and alliinase which extracted from nature garlic. By study the ratio of alliinase catalytic pyrolysis alliin, to establish the basis of alliin/alliinase preparation. To research the stability of allicin solution by HPLC and to investigate the influencing factor. By using HPLC-MS analyze the degradation products and the impurities of the allicin solution to improve and to optimize the technology of the biosynthesis of allicin. Methods: 1. Using HPLC to determine the reaction product by considering both of the dose and the cost. 2. By consulting the method from the European Pharmacopoeia to study the stability of allicin in three temperature requirement and to calculate the half life of allicin as the standard of the evaluation of allicin stability. 3. To establish the analytical method of the allicin solution and the alliin by HPLC-MS and to resolve and identify the structures. Results: 1. Ratio of alliin/alliinase in the technology of the biosynthesis of allicin is 1mg alliin requiring 1.3U alliinase. Because alliinase is a suicide enzyme, alliinase is inhibited by the allicin which generated in the reaction system. The temperature of this reaction is 25℃and to filter the macromolecular substance by membrane separation technique. 2. The influencing factors of allicin stability are high temperature and oxygen in atmosphere. 3. The impurity of allicin solution is methylacylthiosulfatide. The allicin solutions synthesised by different contents of alliin contained the isomeride of alliin and allicin. Conclusion: This thesis optimized the technological conditions of the biosynthesis of allicin, investigated the stability of allicin solution, analyzed the source of the impurity of allicin solution and determined the structures of compounds. The research provided reliable evidence and important foundation for the research and development of the new garlic preparations and the submission of the new drugs.
