This thesis includes three chapters. The first chapter summarized the most recent advances in the research of the chemical constituents of tobacco leaf. The second chapter presented the studies on the chemical constituents of the safflower jinyuan tobacco, which extracted by 70% methanol-water, as well as identifications of all the structures. The third chapter analysized the volatile aroma components of tobacco leaf by means of GC/MS.The review part summarized the chemical constituents of tobacco (Nicotiana), including studies on volatile aroma components.In the research of chemical constituents part,39 compounds were isolated from safflower jinyuan tobacco leaf via a variety of modern chromatographic separation and purification technology. All structures were identified and confirmed by means of the determination of physical and chemical constants (IR, UV, CD, NMR, HRESI-MS) and spectral data (1D/2D NMR).Two phenolic compounds,14 sesquiterpene compounds,2 nishimatsu alkanes diterpenes,6 flavonoids compounds,6 isocoumarins,5 sterols and 2 triterpene compounds, together with 2 other types were isolated from safflower jinyuan tobacco. Those compounds were named a-Tocopherol (1), a-Tocopherylquinone (2), Nicosesquiterpene A (3*), Nicosesquiterpene B (4*), Nicotianasesterpene A (5*), Nicotianasesterpene B (6*),2-keto-a-cyperone (7), Ligudentatol (8), capsidiol (9), 1-β-hydroxy-a-cyperone (10), arundinol B (11), cyperusol C (12), capillosanane Z (13), capillosanane Y (14), curcumanolide B (15), procurcumenol (16), (1S,2E,4S,6E, 8R, 11S)-8,11-epoxy-2,6,12 (20)-thunbergatrien-4-ol (17), (1S, 2E,4D,6E,8R,11S, 12E)-8,11-epoxy-2,6,12-thunbergatriene-4-ol (18), 4’,6-dihydroxy-7,8-dimethoxyisoflavone (19*), 8-acetyl-4’-hydroxy-7-methoxy-6-methylfavone (20*), licoisoflavone (21), 3,5,7,4’-tetrahydroxy-coumaronochromone (22), rugosaflavonoid B (23), catechin (24), tabaisocoumarins D (25*), tabaisocoumarins E (26*), tabaisocoumarin A (27), caudacoumarin B (28),3-acetoxyl-8-hydroxyl-isocoumarin (29), (10R)-7-hydroxy-3-(2-hydroxy-propyl)-5,6-dimethylisochromen-l-one (30),β-sitosterol (31), stimasta-5,11-dien-3β-ol (32), Obtusifoliol (33), Cycloeucalenol (34), Citrostadienol (35), β-amyrin (36), cycloeucalenol (37), Dihydroactinidiolide (38), 1-acetyl-β-carboline alkaloid (39), respectively. And (3), (4), (5), (6), (19), (20), (25), (26) are new compounds. Biological activity testing shows that they have good resistance to tobacco Mosaic virus (TMV) biological activity.The volatile composition which was extracted from the leaf of safflower jinyuan tobacco was analysized by gas chromatography tandem mass spectrometry (GC-MS) using semi-quantitative analysis. The phenyl ethyl acetate was used as an internal standard. The content of the compounds was calculated according to peak area ratio and concentration ratio of a certain compounds and internal standard substance.
