Near-field scanning optical microscope can break through the diffraction limit, which the classic optical microscope can't breach. Its high-resolution, non-invasiveness and no breakage ensure that it will have a prominent role in the future.It can operate at ambient conditions or in a fluid environment and it were widly used in the study of the nano-scale optical imaging and the nano-scale spectrum.Based on the first generation Photon Scanning Tunneling Microscope(PSTM),the atomic force/photon scanning tunneling microscope(AF/PSTM) eliminate the optical spurious image with the method ofπsymmetry two beams lighting, it can be used in many fields and is signality in biomedicine and nano science and technology development.But now the lighting of the AF/PSTM is difficult to arrange in the experiment, which is bad for the commercial of AF/PSTM and the detection of samples with short live time as live cells. A new lighting is promoted, and it needn't to arrange over again every time the probe is changed. It will greatly reduce the difficulty of its operation.And according to the application requirements of the the new lighting, a high numerical immersion objective is designed, and the design of the objective is introduced in detail, and the analysis and estimate of the image quality is also generally given, the result of design shows the NA of this objective is about 1.55, fulfills the requirements of the new illumination system well.The key element of AF/PSTM is the bi-funtional bent optical probe, which is used to collect the signal of atomic force microscope and photon tunneling microscope. When the distance between probe and the lighted sample is settled, the aperture size of the probe which the lights permeate decides the resolution of the system. So, the bi-funtional bent optical probe is vital in the detection of AF/PSTM. The probe is made from normal cheap communication optical fibers, which is fabricated using mechanically drawn and chemical etching, the apex diameter of the fiber probe was less than 100nm.The method to fabricated the bent probe is summarized in a round, including the choice of the experiment parameters, the control of the chemical eching time.
