Phllodendron amurense Rupr.is one of the three hard-leaved species in Heilongjiang Province, because of its medicinal value and double timber value. Phllodendron amurense is under deforestation, resulting in sharp drop in the amount of resources. Recently, Phllodendron amurense,which has been classified as a rare species, national-level protected species and national level 2 protect trees, mainly distributed in Heilongjiang Province in the mountainous south Xiaoxing’anling. Because of the slow growth, Phllodendron amurense resource recovery is extremely difficult. The present study is collected through surveys and germplasm of Phllodendron amurense to carry propagation technique research, establish Phllodendron amurense provenance, and pedigree gene collection zone, which will lay a solid foundation for the Phllodendron amurense genetic improvement and germplasm innovation, to improve the breeding efficiency of Phllodendron amurense, to promote the sustainable development of great significance.1. The germplasm collection and excellent provenance, family selection of Phllodendron amurenseThe research carried out on selection standard of Phllodendron amurense plus trees. The selection criteria of Phllodendron amurense plus trees are the trees with symmetry crown, thin collateral, narrow crown, strong apical dominance, good natural pruning, no pests, no mechanical damage, no large dead knots and dieback, no forking phenomenon, normal strong energy, rapid growth and tall body. Quantitative indicators as standard are that the average height or diameter of plus trees should be 1/2 times of the standard deviation greater than the average height or diameter of forest stand. For seeds from four kinds of sources to carry out seed traits variability of study results:The provenance seeds length, seeds aspect ratio of different Phllodendron amurense are significantly different, are present variation, seed width do not have significantly difference; the provenances seed quality of different Phllodendron amurense exist pole significant variation, the kernel mass of Linkou provenance seed is the largest, while the kernel mass of Harbin provenance seed is minimized. The present study carried out seedling breeding 1-year-old or 2-year-old seedling variability of growth, results showed:The best planting period of Phllodendron amurense in Heilongjiang Province is from 15th to 25th September; the best seed treatment is using frozen processing, when Phllodendron amurense seedlings sown in the amount of ridge culture or bed culture, the sowing coefficient is 4.0, the excellent seedling density of Phllodendron amurense is 60 to 70/m2 in the ridge culture,80 to 90/m2 in the bed culture, directional breeding seedlings should choose S2-0 type Ⅰ,Ⅱ grade seedlings. There are no significant differences in the height growth and diameter growth of amurense different provenances progeny 1 year old seedlings; Shuangyashan provenance progeny diameter growth is relatively good. The height growth of Phllodendron amurense different provenances progeny 2-year-old seedlings has significant differences, while no significant difference in diameter growth. Xinglong, Harbin, Shuangyashan provenance progeny height, diameter growth is relatively good, Lin6, Lin7, Lin8, Xingl, Xing2 progeny seedling growth is more prominent, which can be protected as an excellent genetic resources and use. In addition, the research carried out 10-year-old Phllodendron amurense provenance, pedigree variation and selection, the results showed that:10-year-old Phllodendron amurense has three provenance trial forests are growing well, different provenances grown significantly different, Linkou provenance is better. Within three kinds of sources, exist abundant variations. Dailing provenance’s height, diameter variation are the largest.17 Phllodendron amurense pedigrees have significant different height and diameter. The top 5 pedigrees of height and diameter are Linkou provenance, related to the local provenance.2. The asexual propagation technology research of Phllodendron amurense The study conducted the asexual propagation technology research of Phllodendron amurense, including tissue culture, grafting, cutting etc..The results show:Drawn from different parts of the bud induction frequency is not the same, hypocotyl is most likely to be induced, followed by shoot tip and cotyledon, stems and leaves are more difficult to be induced. Clef grafting is the best way of Phllodendron amurense grafting, the best time of grafting is from late April to early May, the appropriate height of grafted rootstock are 6 to 8cm. The appropriate time of Phllodendron amurense tender branch cutting is in late July, the best cultivation medium is vermiculite, the test range IBA500ppm, NAA each concentration is better for roots of Phllodendron amurense.3. The gene preservation forest construction techniques of Phllodendron amurenseThe study carried out the construction techniques of Phllodendron amurense gene preservation forest. The results show:The gene preservation forest construction needs knowing the position, site conditions and confirming reasonable density, seeding method and tending measures.2 gene preservation forests are constructed, occuping 2.6 hectares.
