Euonymus maackill Rupr,euonymus euonymus,deciduous shrub or small tree,because of its high ornamental value,practicality,and can also be used as medicine,it is deeply loved by people.At present,Euonymus maackill Rupr is mainly propagated by sowing and cutting.The growth period of seedlings is long,and it is prone to pests and diseases.The offspring are also prone to mutation,and it is difficult to maintain the excellent traits of the parents.Using cuttings to breed,the seedlings are difficult to root and the roots are shallow,the seedlings grow weak,and the wounds are susceptible to bacterial,fungal and microbial contamination,which causes the cuttings to accumulate a large number of pathogens,resulting in slow growth of seedlings and even death.It restricts the large-scale production of Euonymus maackill Rupr,and it is difficult to promote and apply it,which is far from meeting people’s needs.With the development and continuous improvement of agricultural biotechnology,many scholars have carried out different levels of research on these issues.Plant tissue culture technology,as the most complete and widely used plant biotechnology,has been applied in many plants.The use of tissue culture technology can not only achieve rapid propagation of seedlings,but also obtain more quality seedlings.In order to obtain more sterile seedlings and improve the reproductive efficiency of tissue culture,it is usually necessary to add one or more antibiotics to detoxify the seedlings,improve the quality of tissue culture seedlings,enhance the stress resistance of seedlings,and improve the technical level of seedling production.It satisfies the demand for Euonymus maackill Rupr seedlings in production,and also reduces the use of pesticides,which is of great significance to environmental protection.In this experiment,Euonymus maackill Rupr annual shoots with axillary bud stems were used as test materials.Through the study on disinfection treatment of Euonymus maackill Rupr explants,lateral bud induction germination,lateral bud detoxification treatment,lateral bud proliferation culture and induced rooting,it was explored to be suitable for silk cotton planting.The best method for body detoxification and rapid propagation culture provides theoretical and technical support for the development and application of Euonymus maackill Rupr and the large-scale breeding of seedlings.The following are the main findings:1.Different concentrations of sodium hypochlorite(containing 2%available chlorine)were used to disinfect the Euonymus maackill Rupr stem explants at different times.The comparati-ve analysis showed that the optmal disinfection methods of the Euonymus maackill Rupr stem was 70%alcohol for 30 s and 30%sodium hypochlorite solution(containing 2%available chlorine)for 10min.The pollution rate was 16.11%,the mortality rate was 36.44%,and the survival rate was 47.45%.2.The germination of Euonymus maackill Rupr lateral buds was induced by different osmotic pressures and different concentrations of gibberellin.By comparison and analysis,the best conditions for the lateral bud induction germination of suitable Euonymus maackill Rupr explants were MS+6 mg·L-1Gibberellin A3+30 g·L-1 sucrose+7 g·L-1 agar,the number of side buds was 28,and the germination rate was 84.78%.3.The Euonymus maackill Rupr side buds were detoxified by different treatments of nystatin,gentamicin and carbenicillin.The best way to remove side buds by comparison and analysis was to add to MS medium 400 mg·L-1carbenicillin+80 mg·L-1 gentamicin+150mg·L-1 nystatin combination of three antibiotics,the infection rate is 0%;the mortality rate is6.56%;the survival rate is 93.44%.The degerming effect is the best,and the Euonymus maackill Rupr side bud endophytic bacteria have a good inhibitory effect,and the lateral bud growth potential is strong.4.The experiment used different concentrations of 6-BA,IAA and NAA to proliferate and culture the silk buds of silk cotton,and compare the different concentrations of 6-BA+IAA and 6-BA+NAA to treat the proliferation of silk buds.The coefficient and the average plant height of the lateral buds were compared.The treatment combination for the proliferation of silk buds was MS+0.2 mg·L-1 6-BA+0.4 mg·L-1 NAA.The lateral buds had strong growth potential and proliferation.The coefficients and average plant height were the highest,being5.47 and 3.20 cm,respectively.5.The experiment used different osmotic pressure and different concentrations of NAA,IBA combination treatment to induce rooting treatment of Euonymus maackill Rupr group seedlings,through comparison and analysis,the best treatment combination for Euonymus maackill Rupr group seedling induction rooting was:1/2MS+0.4 mg·L-1IBA,the tissue culture seedlings had the highest number of roots and strong rooting,the average number of roots per plant was 3.0,and the rooting rate was 90.77%. |
