In order to make a full play of the excellent petformance of Boer goat at meat production, and merits of the native breeds of Jiangsu in durance and adaptation of herd conditions so as to breed a new strain of meat goats in a shoit time, AFLP (amplified fragment length polymorphism) and RAPD (random amplified polymorphism DNA) techniques were applied to analyzing the genetic diversity of 116 goats from three bieeds (namely Boer, Xuhuai White and Haimen goat), and also the relationship between RAPD markers and growth ,development and reproduction of Boer billy goats. The aims of the present study were: o appraise the advantages of AFLP in researches of genetic diversity of goats compared with RAPD technique. o seek RAPD markers relative to the body weights and body sizes of Boer goat, and also RAPD markers relative to the reproductive ability of Boar billy goat, which were useful in researches of gene marker assisted selection, gene localition and mapping in goat breeding. Moreover, this was in favour of realizing the earlier selection of economic properties and speeding the breeding of new strains of meat-using goats in the country. o develop measurements for better using and protecting native breed resources of goats in Jiangsu. 1. Analysis of Boer goat.Xuhuai White goat and Haimen goat using AFLP and RAPD Goats for experiments were from three breeds, including 60 Bocr goats (20 blood samples and 40 tissue samples and the amount of billy goats and nanny goats equal), 30 Xuhuai goats (1 blood sample from each goat and the amount of billy goats and nanny goats equal) and 15 Haimen goats (1 blood sample from each goat, 7 billy goats and 8 nanny goats). Extracted DNA from these three breeds and constructed DNA pools respectively. To analyze the AFLP polymorphism of the three breeds, 36 selective primer combinations were utilized,29 of which could produce polymorphism. Markers produced by the amplification of these 29 combinations totaled to 3253,including 92 variable markers. Then each combination had 3.17 variable markers at average. The polymorphic frequency was 2.8%. The combinations capable of producing more variable markers included: E00+ACGIMOO+CAC (13) , E00+ACG /M00+CAG (10) ,E00-I-AACIMOO+CAC (8) and E00+AACIMOO+ACT (7) . To get primers of higher polymorphism arid repeatability, 22 primers were selected from 93 arbitrary primers to amp1if~ three DNA pools and 183 markers were gotten,averaged 2.73 markers per primer. The polymorphic frequency was 32.8%. Statistically the two techniques got the same conclusion that in terms of genetic distance, the relationship between Xuhuai goat and Haimen goat was nearer, Xuhuai goat and I3oer goat next to it, while Haimcn goat and Boer goat the furthest. So according to IJPGMA,Haimen goat and Xuhuai goat were classified into one group, and Boar goat the other. Both two techniques could embody the genetic difference among the three breeds effectively. However,A FL? could produce more variable markers, so it was more effective than RAPD. 2.The relationship between body weights and sizes of Boar goat and R.APD markers 20 billy goat tissue DNA samples from Boar goats aged 5-6 month were amplified by 19 selected primers and 156 markers were gotten, including 47 variable markers. The results oft test suggested that OPHO7.4, OPHI3.6, OPH13.8, OPI-115.6, OPEI7.4 arid OPE18.8 had significant effects on weights at weaning and average daily gain during weaning stage.
