The Study On The Relation Between Structure And Development In Culture Condition Of Different Buds From Paeonia Rockii T.Hong Et J.J.Li

This research has been done with different buds of Paeonia rockii in the following aspects, such as morphological characture and anatomic structure ,method of sterilization in tissue culturing,state of developing in the same condition,effects on underground bud with low temperature treatment in different length time and GA3 in different density as well as inducement on tissue culture.The work found that there are different not only in morphological character and anatomic structure of the different buds which collected in same time but also in tissue culture.The different density of HgCL and treatment of different time have different effects for different buds.The development rate of underground buds which dormancy was completely disrupted is basically same.The different density of GA3 has no significant effect in growing up of underground buds The germination rate and development rate are completely different with different length time treatment of low temperature ,the result indicates that 720H has the most effective. The calli have been inducing on MS medium added different density of BA and NAA with different organs.The result have indicated that MS +BA1 .5mg/L+NAA2mg/L is the most suitable for forming cullus.The underground bud is the best among different explant petiole and blade are less than it, the induction rate of callus are 100% for underground bud,60% for petiole,30.8% for blade. Finally, the evolutionary level of P.rockii has been discussed on the basis of above mentioned research.
