Soybean is a kind of important economic crop. Heilongjiang province is the important soybean production region. The production of soybean has been affected greatly owing to low temperature. We transformed Choline phosphotransferase gene (cpt) into soybean in order to solving the problem of cold-resistance of soybean. Cold resistance in the low temperature-induced plants is closely related to the synthesis metabolism path of phospholipids. Cholinephosphotransferase is a key enzyme in the synthesis metabolism phospholipids. Phosphotidycholine (PC) is the enzyme product of CPT gene and it is the key component of the membrane. It is closely related to the mobility of membrane and the cold resistance. In this experiment, plasmid PRDH4O 1, which carrying the 35s-35s-AMV-cpt-NOST fusion gene, was transferred into 5 varieties of soybean by means of the pollen tube pathway. We got the Dl generation plants in the next year. Through the selection of kanamycin and low temperature, several transformed seedlings were obtained. PCR of the transformed soybean DNA indicated the integration of cpt gene into the transgenic soybean genome. Southern blotting demonstrated that the cpt gene was indeed integrated into? the genomes of 2 soybean varieties.
