In this experiment the characters of biochemical substance accumulation and the optimum time to harvest of 20 varieties to be chosen as vegetable soybean were studied with biochemical motheds through data analysis. According to the comprehensive estimate in agronomic traits, nutrient quality and appearance quality of 20 accessions, 6 varieties were chosen tentatively as vegetable soybean fit for cultivation in Hebei province. 20 accessions tested and 10 varieties appended were studied on genetic diversity using amplified fragment length polimorphism (AFLP) fingerprinting technique, and the aim was to explore the feasibility of AFLP mollecular marker technique adapted on genetic diversity in vegetable soybean. The main results were as follows:1. The accmulation of fat appeared mainly when the seed began to grow and was sooner than that of protein which appeared at the latter phase. The quick accumulation in amino acid appeared sooner than proein and later than fat, at the latter stage of seed fullment amino acid turned into protein with its content began to reduce. The accumulations of starch and soulable sugar were mainly at the earlier stage of seed growth and then began to decrease from the middle and latter period of the seed fullment. Content of vitamin C grew quickly during earlier stage of seed fullment and then reduced gradually. The optimum time to harvest was determined as during 18-25d after florence.2. Six varieties of soybean with better qualities in maturation, appearance and nutrient were chosen as vegetable soybean which fit for cultivation in Hebei province.3. DNA extration method from soybean dry seed needs to be optimized furtherly, so the leaf was still the first choice from which DNA was extracted. The AFLP (amplified fragment length polimorphism) fingerprints of soybean was established using primer pairs of Msel and EcoRI. The average identification rate of eleven primer pairs was61.22%. The identification rate of M-47/E-46 was 93.3% which could discriminate 28 varieties of 30 accessions completely and the identification rate of M-50/E-38 and M-82/E-36 was 80%. Using some specific fingerprint pairs and characteristic bands could identify varieties and accessions tested. The genetic diversity of the vegetable soybeans tested was studied preliminarily at molecular level. The cluster analysis indicated that the most cultivars with similar parentage clustered together, some were not, wherease, others showed higher genetic diversities which clustered singly. It was certain that AFLP fingerprint technique was feasible in the study of vegetable soybean genetic diversity.
