Rice is one of the mainest grains of human, only next to wheat. Half of population about in the world mainly feed on rice. Now, with the development of social economy greater demand was put on world food production due to the decreasing arable lands and the increasing global population. So improving varieties by seeking and creating the new genes and resources of seeds and increasing the rice grain yield, the main staples for a large fragment of the world population, would contribute significantly to cope with the global food crisis. Heterosis application, mainly including three way cross and two way cross, is one of the principal way to increase rice grain yield. Applied for production of hybrid seeds, three way cross adopting three lines, namely the male sterility line, maintenance line and restoring line, generally sets up the two isolated plots, namely the plot of breeding and the plot of hybrid production, which will increase the productioncost of seeds and effect on the purity of sterility line and the quality of seeds because of the confounding and restrogression of maintenance line. Two way cross usually adopting two lines, namely the partial male sterility line and restoring line, is limited in large areal popularize owe to difficultly distincting hybrid seeds from selling seeds. In recent years, two way cross may be applied for large areal production very quickly thanks to the study on the light-temperature sensitive sterility rice. But the light-temperature sensitive sterility rice in large areal production grows certainly latent danger owe to the shift of fertility with the change of light and temperature.The differenc between rice male sterility line and maintenance line was studied in this paper by means of anther culture and pollen culture to provide test foundation for purification and rejuvenescence of the male sterility line and maintenance line on basis of three way cross. At the same time, the potentiality and the difference of pollen maturation were inquired in virto by means of pollen maturation culture of the male sterility line and maintenance line to provide the direction of reference and think for further researching the pollen abortion progress of the male sterility line and the substance foundation of fertility shift and to provide the theoretical foundation and the effective pathway for changing three lines to two lines and further improving the sterility rate and the sterility degree of sterility line to heighten the hybrid purity of F|. Moreover, an ideal and forthright system is provided for transfering exogenous gene into rice in the progress of pollen maturation culture in vitro.Choosing д-32A, д-32B,G46A,G46B,D62A and D62B as subjects, this paper studied the main factors and the difference between the male sterility line and the correspondingmaintenance line in anther culture, pollen culture and pollen maturation culture.First, some factors were studied in anther culture. It was shown that callus induction rate was influenced by genotype, density of anther plated, medium, KT concentration and organic addends in the medium. The callus induction rate of G type was the highest hi the three types, the callus induction rate of maintenance line was higher than that of the corresponding male sterility line, anthers in density 100 were most desirable, and the improval Ne solid medium supplemented with 2,4-D2mg/L, KT0.5mg/L and lactalbumin hydrolysate500mg/L, proline 100mg/L and glutaminelOOmg/L, appeared to be the optimum medium. The differentiation medium was MS supplemented with KT2mg/L, NAA0.5mg/L, lactalbumin hydrolysate 500mg/L, proline 100mg/L and glutamine 100mg/L.Second, in pollen culture studies showed that the culture effect was influenced by genotype, medium, KT concentration and organic addends hi the medium.
