Brassica Parachinensis Bailey is one of the special vegetables in South of China. For its tender quality and especial flavour, it is regarded as the best vegetable. The adaptability of Brassica Parachinensis Bailey is better, and it grows rapidly, so it can be planted in the four seasons, and has a important role in anniversary supply of vegetable, especially in the off-season. However, it is aheterocrossplant, and the rate of natural hybridization is high, so acquiring a pure variety needs 5 or 6 years in the breeding. With the method of anther and pollen culture, it can be got double haploid plant. Therefore, it may greatly shorten the time of purification in the breeding. Using the four varieties of Brassica Parachinensis Bailey as materials, we carried out the experiment of anther and pollen culture in order to quicken its haploid breeding. The results are as follows:1. The induction of embryoid is closely correlation to the material genotype, the inductive frequencies are significant difference in different genotype. In addition, the embryoid induction frequencies of pollen culture are higher than those of anther culture , but the induction frequencies of material have consistency in two kinds of culture.2. Using plant growth inhibition substance (MET, MH) with a propriety concentration treating donor plants in different time (spraying plants in meiosis, dipping anthotaxy in low temperature treatment and adding them into medium), it is helpful to increase the inductive frequencies of embryoid.3. In anther and pollen culture, using the low temperature treating anthotaxy(4 , 1 ~2day) before being inoculated and high temperature culturing (33 1 ~2day) after being inoculated can promote the embryogenesis and increase embryoid rate.4. The embryoid rate is highest when the sucrose concentration is 13% in anther and pollen culture. In addition, by adding 0.5g/L AC in medium can also improve embryoid differentiation and increase inductivity.5. Adding adequate concentration silver nitrate in anther culture can facilitate the embryogenesis, the proper concentration is 100~120mg/L. Adding excessive silver nitrate can restrain embryogenesis.
