It has been a research hotspot in the fields of biology, medicine-pharmacy and foodstuff science in the recent years to develop the edible and medicinal Se-enriched products with low production cost. Study on the utilization of the liquid culture of the Se-enriched edible fungi offers a safe and effective way to develop the Se-enriched products to protect health. Liquid culture of the Se-enriched edible fungi, which makes the mycelia that grow in the cultural medlium added with Se to accumulate Se and transform inorganic Se into organic state which is more easily absorbed by bodies and safer to human health than inorganic Se, not only has many merits such as short production cycle, low production cost and high invertion rate of organic Se, but also can improve the content and quality of the protein, amino and soluble sugar to different degree.Agaricus blazei, as a kind of the rare edible fungi that has great commodity potence, has curative effect on enhancing the immunity of human body, and especially has functions on anticancer, anticoagular and scalming nervers. It was found that the Se content in fruitbodies of Agaricus blazei was low. If we make good of its mycelia to accumulate Se, the merits of Se and Agaricus blazei can be combined, which will promote its medicinal and economical value and result in a brilliant market prospect. So far, not only the culture of Se-accumulating Agaricus blazei was rarely reported, but also the liquid cultural system about its Se-accumulation and the Se-accumulation mechanism have not been studied yet.The liquid shake culture conditions of Se-accumulating Agaricus blaze were selected to study on the effect which different Se resources and their concentration, liquid cultural medium composition and other culture conditions have on the ability of Se-accumulation and invertion of organic Se , at the same time, the Se-enriching and Se-enduring ability of Agaricus blazei were studied. The preliminary discuss about the physiological and biochemical bases of Se-accumulating Agaricus blazei was carried out by studying on the number, size and shape of mycelium, the content of the protein and soluble sugar, the chemical forms of the organic Se, the activities of seven extracellular enzyme and the patterns of two isozyme.The experiment results show that the optimum composition of the cultural medium of Se-accunmulated Agaricus blazei is as follows: 3%corn starch, l%glucose, 0.3% (NH4)2SO4, 0.1%yeast extract, 0.2%KH2PO4, 0.1%MgSO4-7H2O and 10mg/100mL VB,. Strains domesticated in slant with Na2SeO3 lug/mL, 250mL conical bottle contained with 80mL liquid cultural medium which is added with 20ug/mL Na2SeO3, inoculum quantity 10%, pH7.0 , 25 ± 1℃, 80r/min of shake speed and 8 days of cultural period are the optimum liquid shake cultural conditions.It is proved in this study that Agaricus blazei has strong Se-enriching and Se-enduring ability, so it should be a suitable spawns to be developed into Se-enriched products. A regularity about the Se-accumulation of mycelia under liquid culture is found: Se content of mycelia is positively correlated to Se-component added at the range of l~20ug/mL, and here is the regression equation: Y=3.0458X+0.0249, r =0.9999. The results also show that the Se content in the mycelia is not above the upper limit of human body want of Se when the mycelia of Agaricus blazei grow normally in the liquid cultural medium added with Se. By making a summary of experiment results and relevant date, we make sure that eating mycelia of Agaricus blazei cultured in liquid cultural medium added with 20ug/mL Na2SeO3 is beneficial to human health and never do any harm.The Se-accumulation mechanism of Agaricus blazei is thought to be as follows: The mycelia take in inorganic Se and transform it into organic state which can activate some extracellular enzyme and promote the composition of many enzyme, so it can improve the content of nutritional component as protein, inversely the improving content of the protein also promote mycelia to store more Se in protein, whic...
