High pathogenic Avian Influenza Virus (AIV) is list A infectious agent that put the export of poultry meat at high risk. In this article The current and commonly used AIV test techniques such as Agar gel Immune-diffusion test, HI test, virus isolation test, ELISA, Immune-fluorescence test and real time RT-PCR were had analyzed their application in the field of quarantine and inspection, including their advantage and disadvantage. The most suitable method-real time RT-PCR detecting AIV in the field of quarantine and inspection has been highly stressed.The high sensitivity and specificity of real time RT-PCR were shown by The test result, 10~7 of the diluted allantoic fluid infected by AIV can also be detected, and has no cross-reaction with common Avian virus and Avian vaccines, such as low virulent NDV and IB V vaccine.5 days after the broiler has been injected with AIV vaccine, no matter hypodermal or muscular injection, the real time RT-PCR has not found any AIV nucleotide, indicating the use of AIV vaccine has no effect on the test results.The poultry meat export company shall not use AIV vaccine according to the requirements of state administration for quality supervision and quarantine and inspection. The writer has proposed using HI test to monitor the antibody level, and using real time RT-PCR to detect the AIV possibly existed in poultry meat, to reduce the risk of poultry and its product export, promoting the foreign trade.In order to quicken the speed of poultry meat through Customs, the fast, sensitive and specific test method is most needed in the field of quarantine and inspection. The AIV real time RT-PCR can detect all influenza A virus in one step. The data has shown its sensitivity was almost the same as virus isolation using chicken embryos eggs. The real time RT-PCR technique uses computer to analyze the fluorescence signal given in PCR process, monitor all the PCR process any time and do not need any treatment to the multiplied PCR product, and the result stable under the same procedures with high repeatability, can test large amounts of sample. So the real time RT-PCR overcomes the disadvantage of easy contamination of PCR, has high prospect in the field of quarantine and inspection.
