Twenty microsatellite markers coming from poultry genome were selected from GENE-BANK for polymorphism analysis in silkies. Total 180 individuals from two lines BM and BF, which were used as laying line for the analysis . Allele frequencies, polymorphism information content(PIC) and heterozygosity(H) were estimated by the software of cervus version2.0. There were some results in our experiment .First, we have got five multiplex-PCR sets by the 18 pairs of microsatellite markers successfully. In the five sets there were two sets with three markers and three sets with four markers. The results of full-automated electrophoresis were perfect. Second. 18 pairs of microsatellite markers were polymorphism. The average number of alleles was 6.67, polymorphism information content was 0.656 and heterozygosity was 0.658,respectively.SASv6.12 performed the correlation analysis between markers and start laying traits in silkies . The results indicate there were 5 microsatellite markers, which were significantly correlate to start laying traits. Marker MCW0068 and MCW0200 were significantly correlate to the weight of first egg; marker MCW00681 MCW0154, MCW0208 and MCW0128 were significantly correlate to body weight at first egg. Then the different genotypes of the same microsatellite marker were compared by way of Duncan. The conclusion was as follows: The genotype DE, AB and AA of MCW0068, AD and DG of MCW0200 were significantly correlate to the weight of first egg; The genotype CE, DF, AB of MCW0068, AD and CF of MCW0154, CD, BB and EG of MCW0128, FH, DG of MCW0208 were significantly correlate to body weight at first egg.The study can provide some basic references for egg production traits in silkies. The results also provide some credible information for quantitative traist locus study and marker assisted selection (MAS). All these will accelerate the progress of silkies breeding.
