Speciality:genetics,Breeding and production of animalTutor:Liu Mengzhou Professor Master Candidate:Wang CailianTo investigate the genetic variability within breeds(lines) and genetic differentiation between breeds(lines)of pigs,blood protein polymorphisms in six pig breeds(lines): two foreign breeds Yorkshire(28) and Landrace(26),cultivateed lines Huate seriesA(26),B(24),C(22), indigenous breed Hezuo pig(20) were examined .10 blood protein loci:Tf,Pa,Po,Es,Am1,Am2,LDH,Hp,Cp,Hb were detected by means of vertical discontinuous polyacrylamide gel electrophoresis(PAGE). Gene frequency,genetype frequency and Nei's average heterozygosity were calculated.The results were as follows: ⑴Genetic variations were observed inTf,Pa,Po,Es,Am1,Am2,LDH,Hp,Cp. Hb had none variation.⑵There were distinct characters of breeds in the isozyme spectra and polymorphism of LDH of those breeds(lines):Subbands of isozyme of LDH appeared in Huate seriesA,B,C,but Yorkshire,Landrace and Hezuo pig didn't appear subbands .⑶The degree of genetic variability within breeds(lines) estimated by Nei's average heterozygosity for these 9 blood protein loci(except LDH) was reatively lower in C sersies (0.4665)than that in other breeds(lines).⑷Genetic distance between breeds(lines) were obtained using Nei's and Roger's formula to construct a phylogenetic tree. The tree grouped Yorkshire,Landrace,indigenous breed Hezuo pig in one branch, grouped cultivateed lines Huate series A,B,C in another branch.
