Tricholoma matsutake is an ectomycorrhizal basidiomycete, which forms ectomycorrhiza mainly with pinus densiflora. Artifical cultivation of the fruit-body has not been established in a stable condition. And the mycelium grows very slowly. This thesis investigated nutritional conditions of T. matsutake T2 and effects of herbs on the mycelium growth.As far as T2 was concerned, the medium of PDA added 0.5% yeast extract was the best; Solid flat and liquid stationary methods were better than liquid vibration; The observation period of culture was 30 days; The optimal initial pH value was 5.5, and temperature was 22℃.As for T2, starch, corn flour and dextrose among 25 kinds of carbon sources were better, Moreover corn flour was the best; Yeast extract and beef extract among 26 kinds of nitrogen sources were the best. Corn flour, dextrose and yeast extract were chosen as carbon and nitrogen sources. Effects of the three factors on the mycelium growth were studied by orthogonal rotational regression design of the second power of three factors. The regression equation of the mycelium growth was established. Results showed that optimum medium ingredients were 2.00% corn flour, 2.00% dextrose and 0.55% yeast extract.Effects of 80 kinds of herbs on the mycelium growth of T2 were that 50 kinds of herbs among them had inhibitory effect on the mycelium growth with restrain rates above 10% and 26 kinds of herbs had less effect, while only 4 kinds of herbs (Semigaguilegia adoxoides Makino, Amomun kravanh, Houttuynia Cordata Thunb and Triterygium Wilfordii) markedly promoted the mycelium growth with increase rates above 10%( α =0.05). Furthermore, the first two extremely promoted the mycelium growth( a =0.01). These herbs contain some monoterpenoids or poly-cyclic diterpenoids, which are abundant in Pinus. These substances probably were stimulative to the mycelium growth of T2.Moreover, five strains were isolated from different sections tissue of fruits. Results indicated that lamella was the best tissue for isolation and that the modalities of five strains were basically similar with T2 Strain T3 was chosen to identificate by ITS primers and the result was that it belonged to T. matsutake.
