Tricholoma matsutake is Tricholomataceae fungi, it is a kind of rare wild mushroom withnutrition and medicinal. This text with the product from Tricholoma matsutake fermentation forthe study, explore the whitening effect of mycelium polysaccharide, fermented liquid andalcohol extract. Tyrosinase is a key enzyme which can catalyze generate melanin, while theproduct of melanin is responsible for the process of skin blachening. Free radicals is closeconnection with ageing, Hydroxy radicals, a matter produced in vivo, which can generateproducts of oxidative but instable, Oxygen radicals have oxidation effect during the formationprocess of melanin. If plenty of the free radicals produced without efficient cleaning, theprganism will be ageing uninterrupt.The dissertation mainly research the effect of Tricholoma matsutake mycelium on skinwhitening, including the inhibition of it on tyrosinase, antioxygen and B16cells. From theresearch of the effect on the inhibition ratio of the tyrosinase, the effect of B16cells and thecleaning of free radicals, we got the following results.1. Tyrosine as substrates, research the effect of Tricholoma matsutake fermentation on theinhibition of monophenol. The results show that Tricholoma matsutake fermentation all haveinhibitory activities on tyrosinase, inhibitory activity of polysaccharide from Tricholomamatsutake mycelium fraction is higer than others. Concentration that results in50%activity(IC50) lost is1.18μg/mL. Also there is no change of the lay time on monophenoloxidase. Dopa as substrates, research the inhibition about diphenol oxidase of tyrosinase onpolysaccharide from Tricholoma matsutake mycelium. The results show it have strongersuppression, Concentration that results in50%activity(IC50) lost is1.46μg/mL. The inhibitionkinetics analyzed by Hnewaver-Burk plots showed that both reactions are all reversible andhybrid inhibition.2. The effect of submerged cult products on autoxidation. We discover the three fractionsextracted all have inhibitory activities on free radical, inhibitory activity of polysaccharide fromTricholoma matsutake mycelium is higher than others. The clearance rate of polysaccharide ofTricholoma matsutake mycelium for OH and DPPH were up to31.43%and90.06%. Usingthe Thiobarbit uric acid methods, the inhibition ratio on Lipid peroxidation in vitro was87.7%.3. The effect of Tricholoma matsutake fermentation on B16cells. The results show it haveless influence on cell proliferation, and basically have no toxicity to cells, the whitening effect similar to Arbutin. Through the determination of intracellular melanin content and the inhibitoryeffect of intracellular tyrosinase, we discover that polysaccharide of Tricholoma matsutakemycelium have the best inhibiting effect. The inhibition rate were up to51.47%and57.49%.The above-mentioned research polysaccharide of Tricholoma matsutake mycelium have theinhibitory effect on monophenol oxidase of tyrosinase and diphenol oxidase of tyrosinase. It caneliminate free radicals, anti-oxidation, anti-aging, but also have a certain impact on melanomacells. The price of fermentation mycelium from Tricholoma matsutake far below than fruitbody.So it is promising be used in skin protection cosmetics.
