Rapeseed is one of the most important oil crops of Brassica in Chink. Its planting area reached 733 million ha in 2000, and the production of oilseed over 1100 million ton every year. The consumption of rapeseed oil account for 60% of total vegetable oil consumption. Rapeseed is the main edible oil in China.High quality pure seed is important for high yield and high quality. The main seed identification method includes morphological, biochemical and DNA Molecular marker identification.Random amplified polymorphic DNA (RAPD) is one of molecular marker techniques, that have been wildly used in be variety identification. In this experiment, RAPD-PCR technique was used to detect the DNA polymorphism of 5 rape varieties(XiangYou 15, XiangYou 1%, XiangNongyou 571, Za695 and Za814 ). The fowling results were presented in this paper:1. Three methods CTAB method, SDS method and sorbitol method were used to extract genomic DNA from the seedling of XiangYou 15. The DNA samples extracted by these three methods were tested by using Spectrophotometer, agarose gel electrophoresis. The difference in yield, quality of the obtained DNA were compared, and sorbitol method as the best method in the test.
