The experiment were done in Huanan Agricultural University from 2002-2005.Taking the diploid Vitis davidii seeds provided by Agricultural Bureau of Hecheng district in Huaihua city for materaials,we treated seeds by 0.2%, 0.3%, 0.4%different densities of Colchicine,and abtained teraploid Vitis davidii germplasm (Including chimera)first time.The morphological, cytological,biochemical and physiological characteristics of the mutated plantlets were studied systematically in comparison with that of diploid Vitis davidii.The major results are as following:1.Different colchicine concentrations and different treated time have different inducemental effects.At the same conditions of others,we use the water solution of 0.3% colchicine for 48h to induce polypliods of Vitis davidii and brought the best effects. The inducemental ratio was 36.8%.2.Compared with their original diploid plants,the induced tetraploid plants showed that the leave became shorter,thicker,but the leave index became smaller. At the same time the mutate plantlets was deformed and colour of leave became deepper.The internode length is smaller than the average.3.Basing on the microscopic anatomic study, the stomata density of the leaf on the mutated branches become smaller,the guard cells are larger than the average ones.4.Cytology studing suggested:the choromosome number of mutated plantlets was 2n=4x=76,and some mixoploids were obtained.Furthermore,octoploid can be incuced by colchicine.5.From the metabolism character researches,we found that the content of chlorophyll, soluble protein, soluble sugar and Vc increased.6.From the resistance researches,we found that the activity level of the PPO, POD , SOD increased but the content of MDA decreased. And that of Pro increased.
