The tissue culture and plantlet regeneration from seed of Eremophila bignoniifl -ora(benth.)F. Muell.,cotyledon of Eremophila macdonnellii F. Muell and sprout explants which were from cutting plant of Eremophila glabra(R.Br) Ostenf.(sticky green leaf) were studied.The influence factors of plantlet regeneration were studied during the initial culture,bud induction and rooting culture.Through analyzing the basal medium,phyto-hormone,saccharide and activated charcoal,the optimum mediu -m was conducted.The transplantation trial of the test-tube plantlet which derived from the bacteria-free seedling of Eremophila bignoniiflora(benth.) F. Muell. was made at last. The main results of this paper were conducted as follows:MS was the best basal medium in seed germination of Eremophila bignoniiflora (benth.)F. Muell. and cotyledon induction of Eremophila macdonnellii F. Muell..The initial medium of them were the same:MS+BA1.0mg/L+NAA0.1mg/L.B5 was the best basal medium in initial culture of sprout explants, which were from cutting plant of Eremophila glabra(R.Br) Ostenf. (sticky green leaf). The optimum medium was B5+BA0.5mg/L+NAA0.1mg/L.The induction rate of three explants were 72.70 %, 71.70% and 79.70% respectively.The optimum medium in the bud induction of sprout explants which were from bacteria-free seedling of Eremophila bignoniiflora(benth.) F. Muell. was MS+BAl.O mg/L+NAA0.1 mg/L,and the induction rate reached 73.05% in the medium.The buds were multipled by 5.5 times on 60th days.The optimum combination of factors in bud induction of Eremophila macdonnellii F. Muell. cotyledon was maltose+BA1.0 mg/L+NAA0.1mg/L,and the induction rate reached 72.27% in the medium.The buds were multipled by 5.0 times on 60th days.The optimum combination of phyto-hormone in the bud induction of sprout explants which were from cutting plant of Eremophila glabra(R.Br) Ostenf.(sticky green leaf) was KT0.5mg/L+BA0.5mg/L+ IBA0.05mg/L,and the induction rate reached 67.23% in the medium. The buds were multipled by 4.3 times on 60th days.The best rooting medium of Eremophila bignoniiflora(benth.) F. Muell. was l/2MS+NAA0.5mg/ L+IBA0.05mg/L+l%AC.The rooting rate reached 81.6% in the medium,the average number of roots were 3.9 and the average length of root was 3.1cm on 21th days.In the medium of pearlite: vermiculite: rever sand=3:2:1,the effect on transplant -ation of test-tube plantlet of Eremophila bignoniiflora(benth.) F Muell. was the best,and the survival rate reached over 80%.Moreover,The phenomenon of shoot organogenesis from the cotyledon explant of Eremophila macdonnellii F. Muell. was discovered in tissue culture.
