The Research On Zanthoxylum Ailanthoides Sieb.et Zucc. Of Tissue Culture And Plantlet Regeneration

This paper mainly deals with the study on seedlings(roots,stem and leaf) and yearling'young stem as explants for regenerated plantlets of Zanthoxylumailanthoides Sieb et Zuce.. By means of orthogonal and comparative experiment design,most favorable conditions for plant regeneration were found including basic media, concentrations and types of plant growth regulators and that screening for the various stages of a medium.The origin of adventitious buds and adventitious roots are illuminated through histological observation. The results are follows:1.The explants from seedlings can not used for inducing callus because of disinfectant's HgCl2phytotoxicity and itself very young. However, the explants from yearling'young stem can be used for inducing callus.2.Primary culture:select stem with buds as exlplants able to induced a large number adventitious buds. Disinfector:75% ethanol+0.1% solution of HgC12, surface-sterilized:75% ethanol for 30s,followed by a treatment with 0.1% solution of HgCl2 for 6min.the rate of obtained aseptic explants was 50%.The optimum medium was WPM+ 6-BA2.5 mg/L+NAA 2 mg/L+ IBAlmg/L, and the optimum medium for induction of callus was WPM+ BA 2.5mg/L+NAA1.Omg/L3.Propagate culture:the:theoptimum medium for proliferation are concluded by L9(33) orthogonaland single-factor experiment. The medium WPM+6-BA0.2mg/L +IBA0.1mg/L+NAA0.1 mg/L was propitious to proliferation of buds,the multiplicat-ion rate was 5.6; the shoots longer than 5.5 cm.,and the optimum medium for proliferation of callus was WPM+1.0mg/L6-BA+NAA0.1mg/L+IBA0.5mg/L.BA as the major influencing factors.4.Rooting culture:1/4MS+IBA0.5mg/L was the best rooting medium by L9(33) orthogonaland single-factor experiment., in which the rooting rate was 95%,the average number of roots was 4.6,the average length of root was 4.0cm. Auxin is the essential components and activated carbon no obvious impact on rooting. 5. Hardened and transplanting:transplanting substrate with osteoporosis and maintan power of water that garden surface soil was the best, where the survival rate could reach 96%,the seedlings grew very well.6.Histological observation displayed the adventitious buds and roots of Zantho-xylumai lanthoides Sieb.et Zuce. were both originated in stele.
