Prunus humilis,a species of Cerasus and originate in china,is cold-resistant, able to bear the drought,saline,alkalinly and barrenly, the wild fruit tree with very strong adaptability. The major object of this study is to exploit the technique systems of tissue culture and rapid propagation for Prunus humilis, through the randomised block design, combine analysis of variance, using the stems as the explant, and the optimal medium and culture programe are studied. The major results were as following:1. the explants from apical bud of annual were the best among inoculated seedlings, apical bud of biennial and mature trees ,which had both low contaminated rate,better life and could gain the mature trees fastly, the pollutingrate is 9.62%,livability is 93.61%.2. The middle part in the same plant was the best part of gaining explants, which hadlow contaminated rate and better sprouting effect, the pollutingrate is 10.87%,livability is 95.24%。3. May was the best period of picking the explants in the test and had the best sprouting effect. the pollutingrate is 10%,the rate of browning 8.89%,the death rate is6.67%。4. Temperature for culture: Day 25±2℃,Night 23±2℃.14 to 16 hours illumination every day, and about 1600 to 2000 lx illumination intensity. The pH of medium was adjusted to 5.8~6.25. The best initiation medium was MS+6-BA0.05mg/L+IAA0.1mg/L4g/L.After a month culture, the frenquency of initiation could reach 74.1%.6. The multiplication of shoot clump:MS+ZT0.3mg/L+IAA0.2mg/L+AC2g/L was the best medium.After 30 days culture, the increasing coeffecient was 4.45.7. Stronger nurslings culture: The MS+KT0.1mg/L+NAA0.5mg/Lcould strong nurslings effetively.8. Rooting curture: The 1/2MS+IAA0.7mg/L and sugar 20g/L was the best one in rooting culture. After 2 months culture, the rooting rate is 70%, the average of root of rooted plantlets is 5.93 , and the average root length of them is 4.03 cm .9. the callus induction rate of 97.8% when on MS+KT0.3mg/L+NAA0.5mg/L;Leaf is more suitable as the explant of callus induction than stem,and callus induction is conpact,the color is normal and the protuberance of surface is more uniformity10. the differentiation medium, MS+6-BA0.1mg/L+IAA1.5mg/L was the best to induce buds from callus,the rate is 97.2%,the average of buds is 3.38.11. For transplantion of plantlets, the soil of the rotten leaves in the forest was the best material, and the survial of plantlets could be up to 88% after transplanting soil.
