Discussing on the technology factors which affectingrapid asexual propagation and scale manufacture ofPhalaenopsis.Results showed that:the type of culture medium and thetime of subculture affecting directly the propagation rate ofprotocorm. The propagation rate of protocorm in the modified Kyotoand KC medium is significantly higher than MS medium. The optimalculture time of protocorm is two and a half months. Using the methodof two steps cultivating plantlet woule accelerate itsmaturity,only seven or eight months in tube .According toinvestigate the abloom plant of protocorm of five varieties whichpropagated 20 generations, the results showed that the characterof these blooming plants keep invariable except any specificvarieties. Base on this study, it summarized the operatingprocedure for tissue culture and scale manufacture of Phalaenopsis.Studying on the causes and the recycle effects of vitrificationprotocorm in manufacture of Phalaenopsis.Results shows that:itaggravates the extent of protocorm vitrification with the increaseof hormone concentration and the expansion of subculture time. Thevitrification rates of two culture mediums rise to 71﹪ and 65﹪ respectively when the concentration of hormone NAA is 5mg·L-1 ,hormone BA is 10mg·L-1 and subculture the ninth generation. Thereproduction ability and plant regeneration rate is lowerdistinctly than formal protocorm. The phenomenon of vitrificationcould restore significantly after culture 2 or 3 generations in theculture medium without hormone. The differentiation ability ofprotocorm restored and the state of plant growth is consistent withthe formal protocorm. It can continue applying to the manufactureof tissue culture.
