The short-stemmed stocks of sweet cherry (Cerasus avium Moench) Gisela7# and Rus-25 were used to study the growth phase of explants for choice to inoculate, the multiple of sub propagation as well as the rate of striking root.The results were as follows:1. The Establishment of Explants and the Selection of Culture Medium of the Short Stemmed Stocks of Sweet Cherry Gisela7# and Rus-25The tests studied the pollution rate, survival rate and propagation coefficient of explants of Gisela7# and Rus-25 which were inoculated at different periods; the influence on the differentiation and growth of the explants of Gisela7# and Rus-25 by different density of 6-BA; The selection of the most appropriate culture medium for regeneration of plantlet leaves of Gisela7# and Rus-25 which were induced by different density of 6-BA, IAA, NAA and GA3 . Results showed that the appropriate explants were axillary buds;The appropriate inoculated period was spring and the culture medium MS+6-BA0.5-0.7mg·L-1 was suitable for Gisela7# producing adventitious buds.The highest induction rate was 52.0% and bud seedlings were healthy and strong;The medium MS with 0.6 mg·L-16-BA was more appropriate for induction of the Gisela7# producing adventitious buds.The induction rate was 43.3%, the bud seedlings and the color of leaves were normal;The most appropriate medium for inducing plantlet leaves of Gisela7# and Rus-25 to regenerate were MS +6-BA 2.0mg·L-1+IBA,1.0 mg·L-1+NAA1.0mg·L-1 and MS +6-BA1.5mg·L-1 +IBA0.5 mg·L-1+NAA0.2mg·L-1 respectively. The induction rate was high when the tender leaves of Gisela7# spreading out freshly (until two slices) used as explants.The regeneration rate of Rus-25 was 100% and the average of axillary buds was 6.1; The regeneration rate of Gesila7 was 70.3% and the average of axillary buds was 5.4.2. The Selection of Foundational Culture Medium of Subculture for Short Stemmed Stocks of Sweet Cherry Gisela7# and Rus-25The tests studied the influence on subculture of the Gisela7# and Rus-25 by different density of 6-BA; the influence on subculture of Rus-25 by different treatment of foundational culture medium, 6-BA and ZT; and influence on subculture of Rus-25 by the size of materials. Results showed that the appropriate foundational culture medium for subculture of Rus-25 and Gisela7# was MS; MS+6-BA2.0mg·L-1+NAA0.02mg·L-1 was appropriate for Rus-25. The propagation coefficient reached 4.5 and the rate of effective seedlings was 96%.But the bud seedlings grew long, thin and weak, and the color was yellow. we should choose the optimized mediumt to culture strong seedling. MS +6-BA2.0mg·L-1 + NAA 0.01 mg·L-1 was appropriate for inducing the bud propagation of Gisela7#.The propagation coefficient reached 5.5 and the bud seedlings were healthy and strong.
