The Rhododendron lapponicum has the characteristics of rich colors,beautiful flowers,and beautiful trees.It has high ornamental value and research value,and as a horticultural variety it was widely used in garden planting and tourism development."late flower"Rhododendron is different from others because of its late flowering period.It plays a role in extending the viewing time and promoting the development of tourism in the tourism industry.However,the number of"late flower"Rhododendron in the Azalea is relatively small and the distribution is not concentrated.Therefore,the rapid propagation and cultivation of the"late flower"Rhododendron provide a theoretical basis for enriching the resources of Rhododendron lapponicum and provides a certain technical support for the industrial production of the"late flower"Rhododendron.The study took five species"late flower"Rhododendron?Rhododendron maculatum,Rhododendron decorum,Rhododendron annae,Rhododendron stamineum and Rhododendron jiulongshanense?as research objects.The research was conducted on Rhododendron seed propagation,cutting propagation,and tissue culture,and a set of efficient and rapid propagation techniques for Rhododendron lapponicum were summarized.Mainly get the following results:1.Treatment with different concentrations of GA3 has an effect on the germination of Rhododendron lapponicum.At a certain concentration,the germination index,germination potential and germination rate of the five Rhododendron seeds were significantly higher than those of the control,and the germination time lag,germination peak period and sustained germination lasting time were relatively shorter than those of the control.Among them,each germination indicators of the seeds of Rhododendron stamineum,Rhododendron annae and Rhododendron jiulongshanense were relatively higher when treated with 600 mg·L-1,and the germination rate of Rhododendron stamineum seeds was relatively good when soaked with 700 mg·L-11 GA3.The germination rate of Rhododendron maculatum was higher when treated with 400 mg·L-11 and 700 mg·L-11 GA3.Compared with the germination ability of five Rhododendron seeds,the germination period of Rhododendron maculatum,Rhododendron stamineum and Rhododendron jiulongshanense were relatively short,and the germination index,germination potential,germination rate and seedling rate were higher than those of other two Rhododendrons.2.Different substrates and different hormone ratios have certain effects on cuttings of Rhododendron lapponicum.When the matrix ratio was 4:1,the cutting bud rate of the two Rhododendron is significantly higher than that of the control,which is 33.33%and 63.33%respectively,and with the increase of the content of peat soil in the substrate,the cuttings rate increases first and then decreases the trend of.The bud rate of Rhododendron annae cuttings in the hormone-treated group was higher than that of the control,but the results of the treatments were not significantly different;Treated with a mixed solution of 200 mg·L-1IBA+100 mg·L-1NAA,the Rhododendron stamineum cutting bud rate was significantly higher than that of the control.3.The disinfectant 75%C2H6O,10%NaCl O and 0.1%HgCl2 have different disinfection effects on explants.With the same sterilization time,the pollution rate of the explants after treatment with the three disinfection solutions in turn were:10%NaClO>75%C2H6O>0.1%HgCl2;the survival rate was:0.1%HgCl2>10%NaClO>75%C2H6O.And with the increase of sterilization time,the pollution rate and survival rate of the explants gradually decreased with the three solutions.When disinfecting with 0.1%HgCl2 in the three treatment solutions,the pollution rate was the lowest and the survival rate was the highest,so it was the best treatment solution of explant.4.Among the four explants of leaf buds,flower buds,sepals and stems,the induction rate of callus induction was higher and the browning rate is low when Rhododendron maculatum,Rhododendron stamineum and Rhododendron jiulongshanense used sepals as explants;while the Rhododendron decorum and Rhododendron annae got the same effect when the flower buds are used as explants.Overall,the callus induction rate of Rhododendron jiulongshanense among the five was relatively highest,because all of the four explants could be used for callus induction.However,the callus induction rate of Rhododendron annae is relatively low and the browning rate is high with any of the four explants.5.In the callus induction medium used,added in 2.0 mg·L-11 ZT and 2.0mg·L-1NAA,the average callus rate of Rhododendron maculatum was as high as 98.99%;With the medium(1/2WPM+1.0 mg·L-1ZT+1.5 mg·L-1NAA),Rhododendron decorum had an average callus rate of 90.93%;Rhododendron annae,the hormone concentration at 2.0 mg·L-1 ZT and1.5 mg·L-1NAA,the highest callus induction rate was 51.86%;In addition,with 1.5 mg·L-1ZT+2.0 mg·L-1NAA in the culture medium,the the induction rate of Rhododendron stamineum and Rhododendron julongshanense callus was the highest,which were 94.62%and 96.27%respectively,the growth status of callus in their suitable induction medium is better.6.The sprouting induction medium of the five kinds of"late flower"Rhododendron were different.Among them,the bud induction rates of Rhododendron maculatum,Rhododendron decorum and Rhododendron jiulongshanense were higher in the medium 1/2WPM+3.0 mg·L-1 ZT+1.0mg·L-1NAA),and the bud induction rates of Rhododendron maculatum was 80.00%,the Rhododendron maculatum was 10.00%And the Rhododendron jiulongshanense was 90.00%.Rhododendron decorum induced buds with the ZT and NAA concentration of 1.0 mg·L-1and 0.1mg·L-1.The budding induction rate was 85.00%.Rhododendron annae did not germinated in the nine media in the table as budding induction medium.7.The situation of the five species of Rhododendrons is roughly the same in the cluster culture,the results showed that the five kinds of Rhododendrons had more buds when the medium was 1/2WPM+3.0mg·L-11 ZT+0.1mg·L-11 IBA.The value-added multiples were the highest,and the seedlings were the thickest and the best.The effect with 1/2WPM+3.0 mg·L-11 ZT+0.2 mg·L-11 TDZ medium was the second.8.Five kinds of Rhododendrons rooting media require different hormones,the rooting rate of Rhododendron maculatum was higher when the medium was 1/2WPM+1.5 mg·L-1NAA+1.0 mg·L-1IBA;When the culture medium was 1/2WPM+0.5 mg·L-1NAA+3.0 mg·L-1IBA,the rooting induction rate of Rhododendron decorum was relatively high;Rhododendron annae and Rhododendron stamineum both took root in1/2WPM+1.0 mg·L-1NAA+3.0 mg·L-11 IBA medium,but the rooting rates were low;Rhododendron jiulongshanense has the highest rooting rate when the culture medium is 1/2WPM+1.0 mg·L-1NAA+2.0 mg·L-1IBA.9.When the tissue culture seedlings were domesticated by mixing 3:1volume of peat soil and perlite as the substrate,the survival rate of the tissue culture seedlings of the five species of Rhododendron was higher,and the growth state of the seedlings was better.
