Objective:To define the expression of Ezrin and E-cadherin protein in bile duct carcinoma tissues, investigate the effect of Ezrin and E-cadherin in bile duct carcinoma cell differentiation, pathological grading, prognosis, metastasis and clinical pathological characteristics. Research the functions of Ezrin and E-cadherin in bile duct carcinoma developing and lymph node metastasis.Methods:Collected the bile duct cancer specimens from38patients with complete clinical record from July,2007to July,2010in hepatobiliary surgery of Yantai Yuhuangding Hospital, can be used for specimens in30cases, exclused criteria of two operations. At the same time, selected10normal bile duct disease as the control group. Detected expression of ezrin and E-cadherin of cancer tissue using immunohistochemical method, analyzed the tissue expression using a semi-quantitative method. And cleared cut the correlation the expression levelof Ezrin and E-cadherin of patients and tumor stage, lymph node metastasis in statistical analysis.Results:1. In the collected specimens, the positive expression of Ezrin protein was60%, average score of3.10in control normal tissues, bile duct cancer tissues expression were stronger than normal tissues, the positive expression rate was90%, scored was6.96; phase â… /â…¡ bile duct cancer positive expression rate was82%, the average score of4.64, phase â…¢/â…£ positive expression rate was95%, with the average score of8.32; positive expression rate of carcinoma of bile duct with lymph node metastatic was94%, average score of9.11, the positive expression rate of bile duct cancer without lymph node metastasis was85%, the average score of4.09.Those differences of results were statistically significant.2. Compared with the normal bile duct tissues, the E-cadherin protein decreased obviously in tissue of bile duct carcinoma. positive expression rate in normal tissues was90%, the average score of8.10, the positive expression rate of carcinoma of bile duct was76%, average score of5.51; the positive expression rate of E-cadherin in carcinoma of bile duct that accompanied by lymph node metastasis was65%, the average score of3.06, the positive expression rate of bile duct cancer without lymph node metastasis was92%, the average score of6.15. Those results were statistically significant difference. The positive expression rate of E-cadherin in bile duct cancer phase â… /â…¡ was82%, the average score of6.36, positive expression rate in bile duct cancer phase â…¢/â…£ was74%, the average score of4.16, those differences of results were statistically significant.ConclusionIn:1. The expression of Ezrin in bile duct carcinoma is strong, and there is correlation between the expression and clinical stage, lymph node metastasis of bile duct carcinoma.2. E-cadherin in the tissue of bile duct carcinoma in low expression, and there is correlation between the expression and clinical stage, lymph node metastasis of bile duct carcinoma.3. Ezrin and E-cadherin protein can be used as a referenced marker to bile duct carcinoma diagnosis, monitoring and biological markers.
