After the LV-SNAILsiRNA vector was transfected into the HepG2cell line, and cell proliferation was measured by the MTT assay. E-cadherin mRNA and protein expression were examined by quantitative PCR and western blotting, respectively.Results:1) We successfully constructed the LV-SNAILsiRNA lentivirus vector and proved that it can suppress expression of SNAIL gene in HepG2cells (P<0.05vs. control).2) RNA interference of SNAIL by the LV-SNAILsiRNA construct significantly inhibited the growth of HepG2cells, in addition to significantly increasing E-cadherin mRNA and protein expression (P<0.05vs. control)Conclusion:The Snail expression in human HepG2cell line can be effectively inhibited by LV--SNAILsiRNA lentivirus vector. Besides, We can conclusion that the expression of SNAIL may be responsible for the poor proliferation and survival in HepG2cells. The SNAIL signaling pathway may provide a novel therapeutic target for the treatment of HCC.
