The EliVisionTMPlus immunohistochemical technique and hybridization in situ technique are used to detect the expression of Snail, N-cadherin and mRNA from them in Osteosarcoma, Osteochondroma and normal cartilage. Its aim is to search the following aspects. The first, we investigate the expression of Snail, N-cadherin, protein and mRNA in Osteosarcoma, Osteochondroma and normal cartilage and their relations with clinical pathological features through the experiment. The second, we survey the relations among Snail, N-cadherin and mRNA in Osteosarcoma through the experiment. The last, we also try to find out the consistency of expression of Snail, N-cadherin by using immunohistochemisty and hybridization in situ.38cases with Osteosarcoma,20cases with Osteochondroma and20cases with normal cartilage were collected from the Pathology Dep. of the251st PLA hospital ranging from Jan.2004to Jan.2008. The expression of Snail, N-cadherin and mRNA were detected in three tissues by immunohistochemistry technique of EliVisionTM Plus and hybridization in situ technique. The relationships among the three markers and gender, age, Body Regions, Dahin type, Soft tissues metastasis, Ennecking Stage, and lung metastasis were analyzed. The consistency of expression of three tissues by immunohistochemisty and hybridization in situ was analyzed as well.The positive rates of Snail, N-cadherin and mRNA in38cases with Osteosarcoma were81.6%(31/38),78.9%(30/38) and78.9%(30/38),81.6%(31/38); the positive rates of Snail, N-cadherin and mRNA in20cases with Osteochondroma were10%(2/20),15.0%(3/20) and 15.0%(3/20),5.0%(1/20); while the positive rates of Snail, N-cadherin protein and mRNA in20normal cartilage were5.0%(1/20),0%(0/20), and10.0%(2/20),5.0%(1/20). The positive rates of Snail protein and mRNA in Osteosarcoma were significantly higher than that in Osteochondroma and normal cartilage; the positive rate of N-cadherin protein and mRNA, in Osteosarcoma were significantly higher than that in Osteochondroma and normal cartilage, and there were statistical significances. The positive rate of Snail protein and mRNA in Osteosarcoma were higher than that in Osteochondroma and normal cartilage, and also there was statistical significance; the positive rate of N-cadherin and mRNA were higher than that in Osteochondroma and normal cartilage, and it was statistical significance. The overexpression of Snail, N-cadherin protein and mRNA was correlated with Dahin type, Soft tissues metastasis, Ennecking Stage, and lung metastasis and5year survival rate, but it had no correlation with age and gender.The positive cases of Snail, N-cadherin and mRNA in the38Osteosarcoma cases were31,30and30,31respectively. Postive correlation was found through Spearman test between the expression of Snail and its mRNA and N-cadherin and its mRNA.The overexpression of Snail had close relationship with Osteosarcoma, and it might be involved the regulation mechanism of the pre-cancerous cells to cancer cells. Consistency was found between immunohistochemisty and hybridization in situ in detecting Snail and N-cadherin. Snail and N-cadherin mihgt take parts in the occurrence, development, infiltration and metastasis of the1Osteosarcoma, which were important biological markers in the infiltration and metastasis. Co-detection of Snail and N-cadherin might have important significance to evaluate the metastasis and prognosis of the tumors. The Snail could work as a new marker of potential therapeutic target in the laryanl squamous cell carcinoma.
