Objective: By using the model of MSU, using rats as a test modeland using the late medical specialist Zheng Huaixian’s clinical experience prescriptionfor understanding acute gouty arthritis. Finding the solution is done through the externalapplication of Zheng’s xinshangyaojiawei powder. This powder will show the effects ofusing anti-inflammatory and analgesic procedures on acute gouty arthritis. In additionwe will compare our findings with Sanmiao powder to see whether its efficacy is greaterand superior. Thus, these procedures are designed to provide the theoretical referencesfor clinical treatment of acute gouty arthritis. Method: This experiment used80SD rats,all of rats were built model by using MSU ankle injection. After successful completionof the modeling, we randomly divided the seventy-two rats into three groups accordingto their weight: the rats of group A have jiaweixinshangyao powder, the rats of group Bhave Sanmiao powder, and the rats of group C are the model group. Each group hastwenty-four rats(n=24), within each group, the rats are then divided into four groupsaccording to different times:12hours,24hours,3days, and5days, each group of fourhas6rats (n=6), labeled separately with a marker pen. After the modeling to drugintervention treatment, taking the synovial tissue of rats in12h,1d,3d,5d respectively,and observing the rat changes of skin temperature, gait, swelling in all the phase points.Use the ELISA measuring the changes of ICAM-1and HE staining to observe themorphological changes in the synovial tissue in rat. After the end of the experiment, wedisposed all of the dates by using the software of spss20.0. Results:1.Three groups ofthe rats have no significant difference between weight in all the phase points(P>0.05);2.Skin temperature index changes: Skin temperature shows a trend of decline in all thephase points. After12hours treatment, jiaweixinshangyao powder group showedsignificant difference with the model group (P<0.05), while the Sanmiao powder group had no significant difference with the model group(P>0.05), after24hours and3daystreatment, jiaweixinshangyao powder group and Sanmiao powder group had significantdifference with the model group(P<0.01), while, jiaweixinshangyao powder group hadno significant difference with the sanmiao powder group(P>0.05). After5daystreatment, Skin temperature showed no significant difference between these threegroups, that had been close to normal skin temperature.3. ICAM-1content in synovialtissue: after12hours treatment, compared with the the model group, jiaweixinshangyaopowder group and Sanmiao powder group showed a trend of decline in all the phasepoints, and the jiaweixinshangyao powder group had no significant difference with themodel group(P>0.05), after12hours treatment, jiaweixinshangyao powder group hadsignificant difference with Sanmiao powder group(P<0.05), and had a great differencewith the model group(P<0.01), Sanmiao powder group also had a great difference withthe model group, while the content of ICAM-1hit the highest point in this moment.After3days treatment, the experimental outcomes had no difference with12hourstreatment, But the content of model group ICAM-1had a decline compared with24hours treatment. After five days, jiaweixinshangyao powder group had a significantdifference with Sanmiao powder group and the model group(P<0.01), Sanmiao powdergroup had no significant difference with the model group(P>0.05),But the content ofICAM-1had a decline compared with3days treatment.4.Histomorphology observation:After drug intervention, the score of Synovium lesions in rats are falling in differentstages, jiaweixinshanhyao powder group and Sanmiao powder group had significantdifference with the model group (P<0.05).Cartilage damage in24hours is more obviousthan12hours, the decline trend over time, Lesions of integral in jiaweixinshangyaopowder group and Sanmiao powder group had significant difference with the modelgroup’s (P<0.01).Conclusion:Jiaweixinshangyao powder and Sanmiao powder maybehad obvious anti-inflammatory effect for inhibiting the release of ICAM-1, and thetreatment effect of jiaweixinshangyao powder is much better than Sanmiao powder.Perhaps the mechanism is that the medicine controls the expression of the ICAM-1inthe rat,s synovium.
