Objective: To study the expression of nucleostemin and p14ARF(alternative reading frame) and their clinical significance in breast infiltrating ductal carcinoma(BIDC).Methods: One hundred and ten cases of BIDC tissues and the normal tissue adjacent tumor were selected from Yongzhou Central Hospital from September 2009 to September 2011. The immunohistochemistry assay was used to examine the expression of nucleostemin and p14 ARF. The relationship between nucleostemin, p14 ARF and clinicopathological parameters, such as age, tumor size, TNM stage, histological stage, lymph node metastasis, menopausal status and molecular classification, was determined. The correlation between the expression of NS and p14 ARF in BIDC.Results: The positive expression rates of nucleostemin in BIDC tissues was 64.5%, in the adjacent normal tissues was zero. There was significant difference between BIDC tissues and the control(P<0.001). There was no correlation between the expression of nucleostemin and age, tumor size, TNM stage, lymph node metastasis and menopausal status(P>0.05). The positive expression rate of nucleostemin in histological grade I,â…¡and â…¢was 40.0%(8/20), 60.3%(35/58), 87.5%(28/32) respectively. Nucleostemin was related to histological grade in BIDC(P=0.001). There was statistical significance between gradeâ… and â…¢ grade(P<0.001),â…¡grade and â…¢grade(P=0.007),whereas there was no statistical significance between â… grade and â…¡grade(P=0.115). Tumors with nucleostemin positivity showed a higher frequency of ER or p53 positivity(P = 0.031, 0.032 respectively), whereas NS expression status was not correlated with PR and HER2 positivity(P>0.05). The positive expression rate of nucleostemin in Luminal A, Luminal B, over-expression HER2 and triple negative was 58.3%, 64.7%, 56.3%, 94.1% respectively. The positive expression rate of nucleostemin was related to molecular subtypes(P=0.046). The positive expression rate of nucleostemin in triple negative was higher than other molecular subtypes. The positive expression rate of p14ARFin the control and BIDC was 100% and 46.4% respectively. The positive expression rate of p14 ARF in normal breast tissues was significantly higher than in BIDC(P<0.001). There was no relationship between p14 ARF and age, tumor size, and TNM stage and menopausal status(P>0.05). The positive expression rate of p14 ARF in histological gradeâ… ,â…¡and â…¢ was 70.0%(14/20), 46.6%(27/58), 31.3%(10/32) respectively, and there was significant difference among the three groups(P=0.024). There was statistical significance between gradeâ… and grade â…¢(P=0.006), whereas the correlation between gradeâ… and gradeâ…¡(P=0.07), gradeâ…¡and grade â…¢(P=0.158) was not statistically significant. The positive expression rate of p14 ARF in lymph node metastasis was lower than no lymph node metastasis(P<0.001). Tumors with p14 ARF positive showed a lower frequency of HER2 or p53 positive(P=0.029 or <0.001, respectively), whereas p14 ARF expression was not correlated with ER and PR(P>0.05). The positive expression rate of p14 ARF in Luminal A, Luminal B, over-expression HER2 and triple negative was 53.3%, 58.8%, 37.5%, 17.6% respectively. The positive expression rates of p14 ARF was related to molecular subtypes(P =0.039). The positive expression rates of p14 ARF in triple negative was lower than other molecular subtypes. Nucleostemin positivity was negatively correlated with p14 ARF in BIDC.Conclusions: The expression of nucleostemin in BIDC was significantly higher than in normal breast tissues and correlated with histological stage, the expression of ER and p53. The positive expression rate of nucleostemin in triple negative BIDC was higher than other molecular subtypes. The positive expression rate of p14 ARF in BIDC was significantly lower than in normal breast tissues, and correlated with histological grade, lymph node metastasis and the expression of HER2 and p53. Whereas the expression of nucleostemin was significantly correlated with p14 ARF in BIDC.
