Purpose:To explore Dahuang Zhechong Wan promote angiogenesis of experimentalresearch,provide the beneficial reference for the treatment of ASO.And providingexperimental evidengce for the efficacy of Dahuang Zhechong Wan.Material and method:Material:3-4month old Wistar rats of clean grade50, weight (180±20) g, provided by experimental animal center of LiaoningUniversity of Traditional Chinese Medicine.Composition of high fat diet:62.8%basic feed+150g/L cholesterol+20%lard+2g/L propylthiouracil+20g/L sodiumcholate.Rabbit anti mouse anti factor Ⅷ-fluorescein isothiocyanate (FITC)antibody was purchased from Wuhan boster company.Transforming Growth Factor-β(TGF-β,batch number AE99292Ra),Vascular Endothelial GrowthFactor(VEGF,batch number,AE98006Ra) and Enzyme linked immunosorbent assay(ELISA) double antibody sandwich detection kit were purchased from ShanghaiYueyan biological technology company.Methods:Clean feed adaptive feed after oneweek, through Arterial intimal injury plus fat feed method to establish50animalmodel of ASO.The model rats were randomly divided into4groups:The model controlgroup,Dahuang Zhechong Wan low, medium, and high concentration group and10ratsin each group.Another10rats as the normal control group. Using laser Dopplerflowmetry records bilateral hind limb perfusion of each rat’s perfusion beforemodeling and every half a monthafter mmoded.And calculating the ratio of perfusion.Slicing the specimen at theend of experimentsCalculating the average of cell-CD31+by CD31immunofluorescence staining.This approach reflects the density of new bloodcapillariesResults:1. Compared with normal control group, the left hind limb perfusion modeling has declined in each group of rats.The levels of serum cholesterol, triglycerides,low-density lipoprotein were significantly increased.Differences were highlysignificant (P<0.01).2.Compared with the control group,the perfusion of Dahuang Zhechong Wan low,medium, and high concentration group were significantly improved (P<0.01).Serum cholesterol and triglyceride levels were significantlydecreased,differences were highly significant (P<0.01).3.Compared with normal control group, the blood of rats in each group of VEGFand TGF-β elevated. differences were highly significant (P<0.01).After thetreatment of Dahuang Zhechong Wan, each group of serum VEGF, TGF-β also haveimproved,differences were highly significant (P<0.01).Conclusion:1.Abnormalities of the vascular endothelial cells secrete a function is aimportant material basis of ASO.The change of VEGF,TGF-β are closely relatedto the condition of ASO.2.Dahuang Zhechong Wan can adjusting the secretion of vascular endothelialcells to improve physical state of blood flow perfusion and ischemia.It also canadjust VEGF,TGF-βin the blood.For experimental ASO has good therapeutic effect.
