Eczema is a common inflammatory skin disease, caused by a variety of internal and external factors. Currently, Western medicine often used corticosteroids and antihistamines to treat eczema. But it is easy to cause a variety of adverse reactions, and also easy to appear drug resistance and recurrence. Chinese medicine has the advantage of in the treatment of eczema. It is safe and effective, and side effects are small. But the application is usually lotion, it is inconvenient to use. At present, we need to develop new kinds of Chinese traditional medicine preparations to against eczema, which is effective, less side effects and easy to use.According to the ancient literature research and clinical experience prescription, select cnidium, sophora, cortex dictam, cicada, honeysuckle as prescription drugs to make a compound traditional Chinese medicine anti-eczema cream. Study on the cream quality and in vitro and in vivo transdermal drug delivery. This study provides the basis for the clinical application. The main research contents and results are as follows:①Extraction of drug active ingredients:Extract the volatile oil of medicinal herbs by steam distillation. The extraction method is that each 100g medicinal herbs, adding 8 times amount of water, heating reflux extraction 6h to extract volatile oil. And keep the volatile oil saturated aqueous solution as water phase in cream. Filter the decoction of the compound medicinal herbs, concentrate the decoction by rotary film evaporation in 60℃. Each 1g fluid extract equal to 5g original medicines.②Cream preparation:By comparing the appearance, pH, centrifugal/heat/cold stability of 3 kinds of w/o type cream, choose the best combination of ingredients. The proportion of each component:in oil phase, Beeswax 4%, paraffin 4%, white vaseline 4%, glycerol monostearate 9.6%, liquid paraffin wax 20%, span-80 1.6%, volatile oil 8%; in aqueous phase, twain-80 0.8%,0.8%, fluid extract 8%, volatile oil saturated aqueous solution 40%. Put the two phases in 80℃ water bath to make them complete melting and dissolution, and mix the two phases while they are hot. Emulsifying 5 min by homogenizer with 3000r/min, a brown and exquisite cream are made.③ Cream quality research:Study on the quality of the cream from the appearance, particle size, pH, qualitative identification and content determination. The cream is moderate viscosity, smooth and delicate, easy to spread. By the dyeing experiments and conductance value determination, he cream can be determined as a w/o type cream. Measured by laser particle size analyzer, the average particle size of the cream is 172.7nm. Measured by precise acidimeter, the average pH of the cream is 6.87.In the selected medicinal herbs, osthole is mainly active ingredients of Fructus cnidii. Osthole content is high, and it is easy to be detected, so choose osthol as index components of the qualitative identification and content determination. Do qualitative identification by TLC, homemade cream and osthole show the same color fluorescence spots in the same position.. Use high performance liquid chromatography and fluorescence spectrophotometry to manage the content of osthole in the cream. Hplc determination shows that osthol content of the cream is 0.3003%, the fluorescence spectrophotometric determination shows that osthol content of the cream is 0.3013%. The determination results of the two methods confirm each other. Osthol content results of the two methods are accurate and reliable, we can choose the appropriate measurement method according to the specific situation of the samples.④ Transdermal drug delivery research:In vivo and in vitro transdermal experiment, study the distribution of osthol in different layers of skin. The experimental results show that in vivo and in vitro transdermal experiment result is similar. In vivo experiment, with a single transdermal drug delivery, the osthol content of cuticle gradually increases in 2h to 8h, and decreases after 8h. In the dermis of mice, osthol content increases gradually with time. Osthol blood concentration of mice reachs a maximum at about 4h, the blood concentration decreases significantly after 4h, the blood concentration decreases less and tends to be stable after 8h. In vitro experiment, the osthole content of accept liquid increase linearly with time, th delay time is 1.7h. This indicates that osthol through skin required a longer time. From above results, the effective component in the cream can remain in the skin for a long time, this will be helpful to produce a therapeutic effect on eczema. So, the frequency of administrationan can be reduced to two times a day.The experimental results shows that osthole emit strong fluorescence at 391nm when the excitation wavelength is 350nm. It can be used for the quantitative analysis of trace samples containing osthole, such as biological samples.
