Preparation And Anti Chronic Eczema Study Of Saxifrage Cream

Objective: The aim of present study is to investigate the preparation process,quality standard and pharmacodynamics research of Saxifrage cream,and lay the foundation for the further development of innovative drug for the treating eczema.Methods: The effects of ethanol volume fraction,extraction time and solvent dosage on the content of total flavonoids and tannins in Saxifrage were investigated by single factor experiment and orthogonal design.According to the appearance,color and coatability to select the type of cream,optimization of the key components in the prescription by using the Box-Behnken design-response surface method.Finally,the effects of emulsifying temperature,method and time on the stability of the matrix were investigated.To evaluate the appearance of Saxifrage cream,the p H,particle size,loading and microbial limits were measured and examined.The quercetin was identified by TLC,and to determinate the content of total flavonoids and gallic acid by UV and HPLC,respectively.Then,the stability of the preparation was studied.Xylene-induced mouse ear swelling test and carrageenan-induced rat foot swelling test were used to study the anti-inflammatory effect of Saxifrage cream.Histamine-induced guinea pig pruritus was used to study its antipruritic effects.DNCB-induced chronic eczema in mice was used to study its anti-allergic effects,and the levels of serum TNF-?,IL-2,IL-4 and IL-6 were measured by ELISA.Results: The Saxifrage was extracted by alcohol,the ethanol volume fraction was 60%,the amount of solvent was 12 m L/g,the extraction time was 120 min,the extract was extracted with ethyl acetate.O/W matrix was used as the matrix of this experiment,the prescription of oil content was 1%(g/g),the drug loading was 5%(g/g),emulsifier dosage was 5%(g/g).When the emulsification temperature is 75? and the time is 10 min,the external phase is added to the internal phase,the appearance of the cream is good.The appearance,particle size,p H and microbial limit of Saxifrage cream were in accordance with the the 2015 edition of Chinese pharmacopoeia,the TLC,UV and HPLC with high specificity,negative without interference.During the preliminary stability test,the indexes of the samples were in accordance with the regulations.Furthermore,the results of pharmacodynamics indicated that Saxifrage cream(the amount of crude drug was 1.25 g/g,2.50 g/g,5.00 g/g,respectively),significantly reduced the mice ear swelling compared with the model group,and the inhibition rates were 26.67%,48.42%,52.67%,respectively.Moreover,high dose significantly inhibited the increase of foot swelling of rats,1 h,2 h,4 h after inflammation,the inhibition rates were 55.84%,43.07%,46.50%,and significantly improved the itchy threshold of guinea pig,and the mouse ear lesions score and swelling.ELISA assay results demonstrated that Saxifrage cream significantly inhibited the production of inflammatory markers IL-4 and IL-6 cytokines,while improved the TNF-? and IL-2 cytokines.Conclusion: The preparation process of Saxifrage cream was reasonable and feasible,the quality standard was controllable,and its therapeutic effect on eczema maybe related regulating cytokine levels.
