| Object:To investigate the effect of electro-acupuncture (EA) at Shu Mu points of lung and large intestinal on the pharmacokinetics of components from Da-Cheng-Qi decoction (DCQD) in rats with acute pancreatitis (AP).Method:Forty-eight male Sprague-Dawley rats were randomly divided into normal〠modelã€DCQDã€Shu&DCQDã€Mu&DCQD and Shu&Mu&DCQD groups (n=8). AP models were established by retrograde infusion of 3% taurocholic acid into pancreatic duct of all rats but the normal group. EA was applied to Shu&DCQDã€Mu &DCQDã€Shu&Mu&DCQD groups 2 hours after modeling with 40 minutes each time (Shu&DCQD group:EA at bilateral BL-13 and BL-25 for 40min, Mu&DCQD group: EA at bilateral LU-1 and ST-25 for 40min, Shu&Mu&DCQD group:EA at bilateral BL-13 and BL-25 for 20min, then at bilateral LU-1 and ST-25 for 20min), three times with an interval of 6 hours. DCQD group were fixed without other processing at the same time. DCQDã€Shu&DCQDã€Mu&DCQD and Shu&Mu&DCQD groups were given DCQD by gavage 24 hours after modeling. The blood samples were collected through their tail veins at the time of 10ã€20ã€40 minutes and 1ã€2ã€3ã€4ã€6ã€8〠12 hours after the gavage. The concentrations of the components from DCQD in blood samples were measured by liquid chromatograph-mass spectrometer/mass spectrometer (LC-MS/MS), and the pharmacokinetic parameters were calculated by DAS 2.1.Results:â‘ The Cmax of emodin, aloe-emodin and naringenin, as well as the AUC of emodin and naringenin in Mu&DCQD group were higher than those in DCQD group (P<0.05);â‘¡The Cmax of naringenin in Shu&Mu&DCQD group was higher than that in DCQD group, and the Tmax of naringenin in Shu&Mu&DCQD group was shorter than that in DCQD group (P<0.05);â‘¢The Cmax of emodin, aloe-emodin and rhein, as well as the AUC of emodin and aloe-emodin in Mu&DCQD group were higher than those in Shu&Mu&DCQD group, and the Tmax of emodin and rhein in Mu&DCQD group were shorter than those in Shu&Mu&DCQD group (P<0.05);â‘£The Cmax and AUC of emodin in Mu&DCQD group were higher than those in Shu&DCQD group, and the Tmax of emodin in Mu&DCQD group was shorter than that in Shu&DCQD group (P<0.05);⑤The Tmax of rhein in Shu&DCQD group was shorter than that in Shu&Mu&DCQD group (P<0.05).Conlusion:EA at Mu points, Shu and Mu points of lung and large intestinal can promote the pharmacokinetics of components from DCQD in rats with AP, while Mu points has the better effect than the combination of Shu and Mu points. |
