Antitumor Effect Of Ganoderma Lucidum Strain And Its Material Basis

Part I of the 2005 edition "Chinese Pharmacopoeia" containing Ganoderma for the dry carpohole of Ganoderma lucidum(Leyss.ex Fr.)Karst or Ganoderma sinense Zhao, Xu et Zhang. Their dry fruiting body has the effects of anti-tumor, regulating the immue system and extra.There are about 300 species ganoderma known in the world, and more than 110 species of which are known in china. Caused by different species ganoderma of fuiting body of significantly different chemical composition, the effects also shows the great differences. Modern studies have shown that ganoderma contains polysaccharides, peptides, triterpenes, protein, steroid, mannitol, coumarin glycosides, amino acids, alkaloids, organic acids, trace elements and extra. All the compositions have the effects of anti-tumor, regulating the immue system, anti-aging, anti-radiation and other effects. The varies strain and chemical compositions in different species ganoderma significantly affected the pharmacological activity of Ganoderma lucidum.Based on the above, the paper selected 18 strains of Ganoderma lucidum, which are kept in our laboratory and extracted the chemical compositions through the technological methods of water decoction, which are tested anti-tumor effect on Sarcoma 180 bearing mice. Different strains of anti-tumor activity of Ganoderma lucidum were carried out pharmacodynamic screening test. Through the study we obtain an exact anti-tumor effect fungus strains, ZL83. And further studies of ZL83 were carried out in the paper. First, we detect the appearance of characters, the content of polysaccharides and triterpenes according to Chinese Pharmacopoeia 2005 edition; Then we study the immune regulation of water extract, the enhance effectiveness and reduce the toxicity effect with chemotherapy drug cyclophosphamide on Sarcoma 180 bearing mice. Finally, the ZL83 water extract was divided into three parts through extraction, and the part of butanol extraction was divided into six parts through silica gel column chromatography, the chemical constituents are checked the anti-tumor activity on the S180 tumor-bearing mice or testing in vitro by MTT assay. The results show that:1. By Ganoderma screening anti-tumor experiments, only ZL10, ZL11, ZL64, ZL83 have inhibited effect on S180 tumor-bearing mice, the inhibition rate reached 27.87%,40.45%,31.55%,36.48%, According to the material circumstances, physical and chemical conditions, select ZL83 for future study.2. The character and appearance of fruiting bodies of Ganoderma, and the content of polysaccharide, Triterpenoid are in line with the Chinese pharmacopoeia, which polysaccharide content was 0.78%, triterpene content of 0.333%.3. The ZL83 water extract of fruiting bodies have increased immune activity, combined with cyclophosphamide have a synergistic effect, and can reduce the high-dose cyclophosphamide induced toxicity.4. The parts of butanol and ethyl acetate, Separated from the ZL83 fruiting body aqueous extract, has good activity against S180 tumor-bearing mice, the inhibition rate reached 53.75% and 45.29%.5. The components obtained from butanol and ethyl alcohol by Silica gel column chromatography with a good anti-tumor activity in vitro, The IC50 for MGC-803 and LoVo cell lines respectively to 94.0 u g/ml 8 and 53.22μg/ml.In summary, we obtained four kinds of Ganoderma lucidum which have significant anti-tumor effect. In the continued study of ZL83, we confirmed that the strain has good antitumor activity in vivo, and can combine with cyclophosphamide, attenuated synergistic role played by the separation of its chemical composition and anti-tumor activity. it’s indicated that the main anti-tumor substances concentrated in the butanol extract, to formic acid eluted by ethyl and ethanol groups received anti-tumor activity into ZL83 main material component.
