As an essential trace element for human beings, Selenium has important biological functions and has a close relationship with the illness, especially tumor and angiocardiopathy, which can severely endanger the health. Ganoderma Lucidum(GL) have been long known to be good to the health and have the ability of accumulating high levels of trace elements. The products of Se-enriched GL combining the effect of GL to that of selenium have practical values.The extraction of effective components of Se-enriched GL and the study on the effect of tumor inhibition can provide theoretic evidence for the exploitation of products of se-enriched GL.In this paper, the in vitro antioxidation of proteins of different solubility characteristics was studied and the extraction conditions of water-soluble protein were optimized. Furthermore, the effect on tumor inhibition of water-soluble protein was studied through animal experiments.Water-soluble, alcohol-soluble and alkali-soluble selenium-containing proteins were extracted by different solvents. The contents of selenium in the three kinds of protein were detected and the results showed that the water-soluble selenium-containing protein (WSSP) has the highest content of selenium. The studies of the antioxidation in vitro of the three kinds of protein showed that the WSSP has the strongest reducing power, the strongest abilities of free radical scavenging and anti-peroxidation.The extraction technology of WSSP was studied. The conditions determined by single-factor tests were described as follow : the pH value=9 and the extraction was taken at 60℃for 8h, the ratio water of to raw material was 40:1(v:w), then the extraction was repeated twice. The author optimized the extracting parameters according to orthogonal experimental methodology and the results showed that the optima conditions of extraction were: pH value=9, the extraction temperature was 60℃, ration of water to raw material was 35:1. Under these optimized conditions, the extraction rate of soluble selenium-containing protein was 51.56%, in which the content of selenium was 533μg/g.In vivo, the anti-tumor effect of WSSP was evaluated by observation of rate of tumor inhibition, the immunity index, the content of MDA and H2O2 and the enzyme activities of SOD and GSH-Px in the silver and serum of mice. The results suggested that WSSP had obviously inhibited the growth of tumor in vivo (the inhibitory rate at the high dose was 41.96%), increased the weights of mice, decreased the content of MDA and H2O2 and increased the enzyme activities of SOD and GSH-Px. The pathology pictures of tumors and organs showed that there were a lot of apoptosis cells in the group of high dose of WSSP.
