The study was mainly to probe an efficient way through tissues culture and rapid propagation of Perennial chamomile. The stems with axillary buds and the stems without axillary buds were both taken as explants. The factors that affect tissue culture including basic medium and hormones were detailed investigated. As a result, the optimum mediums of tissue culture were screened. A plant regeneration system of Perennial chamomile was estabilished in this research, which provide the theoretical base and technical measures for protection and industrial producing of this wildlife resource. The main results as follows:1. Callus induction of aseptic stems: The aseptic stems withoout axillary buds in vitro can be used for inducing callus, and the optimum medium was MS+BA1.0mg·L-1+NAA0.1 mg·L-1. The initial rate was 90%.2. Proliferating cultivation of callus: The optimum culture medium for proliferating callus was MS+BA1.0 mg·L-1+NAA0.1 mg·L-1, proliferating rate was 93.33%.3. Callus differatiation: The optimum culture medium for differentiating callus was MS+BA1.0 mg·L-1+NAA0.5 mg·L-1, differentiating rate was 82%.4. Induction of axillary buds of stems: The stems with axillary buds of young branches were taken as explants. The optimum culture medium for axillary buds'induction was MS+BA0.5 mg·L-1+NAA0.2 mg·L-1, the induction rate was 92%, bud number was 5.4.5. Rooting culture:The optimum culture medium for rooting was 1/2MS+IAA0.2 mg·L-1. The rooting rate could be up to 100%, the root number was 4.9.6. Proliferating cultivation of roots: The optimum culture medium for proliferating roots was 1/2MS+ IAA0.2 mg·L-1, the root number was 4.93. Under the conditions of variable temperature and in the lights, the optimum medium for rooting was 1/2MS + IAA0.2 mg·L-1, The rooting rate was 100%. Under the conditions of constant temperature and in the lights, 1/2MS + IAA0.2 mg·L-1and 1/3 MS + IAA0.2 mg·L-1 were the optimum mediums for rooting, the rooting rate were both 86%. Under the conditions of constant temperature and in the dark, the optimum medium for rooting was White + IAA0.2 mg·L-1, the rooting rate was 73%. Under the conditions of variable temperature and in the dark, the optimum medium for rooting was White+ IAA0.2 mg·L-1, the rooting rate was 87%. 7. Transplanting and cutting:The optimmu transplanting and cutting medium forperennial chamomile is cinder,in which the survival rate was very high and growing very well.
