Wild Pleurotus Ferulae saprophytic or parasitic Ferula songorica Pall roots, study Pleurotus Ferulae NB-1 and Ferula songorica Pall of materials, isolated ferulae chemical composition by activity guided. Study on different chemical composition on the mycelial growth length of Pleurotus Ferulae, a variety of protective enzymes, and the effect on protein expression.Research the chemical substances in ferulae on mycelial growth of biological effects of Pleurotus Ferulae. The main study results are as follows:(1) Ferula songorica Pall roots, 95% ethanol extract (Extracts) using different extraction solvents are petroleum ether extract (Ex.P), chloroform extract (Ex.T), ethyl acetate extract (Ex.EA), extraction residue (R). Activity guide selection chloroform extract (Ex.T), using silica gel column separation method, activity guide selection Ex.Tc, using silica gel column separation, activity guide to select Ex.Tc.3 identified by mass spectrometry, indicating that Ex.Tc. 3 ,there are seven major chemical constituents.(2) Extracts on Pleurotus Ferulae mycelium growth experiments show that the amount of low-added role in promoting the growth of the mycelium was significantly better than CK, a high concentration showed inhibitory effect on mycelium growth promotion. Ex.P, Ex.T, Ex.EA, R, have shown on the mycelium growth of Pleurotus Ferulae catalytic role, which Ex.T the best. Ex.T using silica gel column separated 12 components, there are 10 separate components demonstrated role in promoting the growth of Pleurotus Ferulae mycelium, of which Ex.Tc the best. Ex.Tc column isolated from six components, role in promoting the growth of the mycelium is superior to CK, which the Ex.Tc.3 the best. Ex.Tc.3 low concentration of the mycelial growth of Pleurotus Ferulae no apparent effect of high concentration of the growth of the mycelium to inhibit.(3) Compared with CK, Ex.T, Ex.Tc, Ex.Tc.3 can improve the Pleurotus Ferulae mycelium CAT, POD, SOD activity. As the medium Ex.T and Ex.Tc added increases, the Pleurotus Ferulae mycelium CAT activity showed a gradual increase in the trend, showing a strong dose effect; Ex.Tc.3 on mycelium CAT activity has a dual effect. As the medium Ex.T added increase, the Pleurotus Ferulae mycelium POD activity showed a gradual increase in the trend, Ex.Tc and Ex.Tc.3 on mycelia POD activity has a dual effect. Ex.T on mycelia of Pleurotus Ferulae SOD activity has a dual effect, with the medium Ex.Tc and Ex.Tc.3 added the increase in the amount of mycelia of Pleurotus Ferulae SOD activity showed a gradual increase in the trend, both between the dose effect. Ex.T, Ex.Tc, Ex.Tc.3 on mycelia of Pleurotus Ferulae PPO activity of the dual role of both demonstrated low concentration can improve the Pleurotus Ferulae mycelium PPO activity, high concentration of the mycelium of PPO activity to inhibit.(4) From the CK in the two-dimensional gel electrophoresis can be clearly isolated from the 826 protein spots, from the Ex-P can be clearly isolated from the 592 protein spots, from the Ex.T-P can be clear separation of the 697 protein spots. CK, and Ex-P total of 397 protein spots matched, matching rate of 55.99%, Ex-P in the 134 protein spots were compared with CK significant increase took place, there are 113 protein spots marked down occurred. CK, and Ex.T-P a total of 571 protein spots matched, matching rate of 74.98%, Ex.T-P there are 170 protein spots were compared with CK significant increase took place, there are 112 protein spots marked down. Ex.T-P and Ex-P total 359 protein spots matched, matching rate of 55.70%, Ex-P has 113 protein spots than Ex.T-P significant increase took place, there are 97 protein spots occurred in significant declines. In the medium to add Extracts Ex.T inhibit mycelia pI 5.0-pI 8.0 protein expression, Extracts can promote the mycelia of pI 4.0-pI 5.0 (acidic) protein expression, Ex.T can promote mycelium pI 7.0-pI 9.0(alkaline) protein expression. Extracts and Ex.T compared with the CK Pleurotus Ferulae mycelium can promote the expression of low molecular weight proteins, while the expression of high molecular weight protein has produced a significant inhibition, Extracts on P.Feruli mycelium small quantity of protein, activation of expression of the role of the most obvious.(5) Differences in protein spots identified by mass spectrometry Spot 748 as the hypothetical protein MPER07859, Spot 1180 for the 1,4-benzoquinone reductase, Spot 1363 as the hypothetical protein NECHADRAFT96302, Spot 920 for the 70 kDa heat shock protein, Spot 933 as the hypothetical protein, Spot 1030 for the flavin-binding monooxygenase, Spot 1347 and Spot 484 can not get in the Mascot Search Database retrieve information.
