By agar diffusion method (punch method) and two-fold dilution method in test tube, in vitro bacteriostasis test about Staphylococcus aureus, Escherichia coli, Streptococcus agalactiae of huanglianpu injection on dairy cow mastitis were investigated. The resuLts showed that the bacteriostasis diameters of huanglianpu injection on Staphylococcus aureus, E. coli, Streptococcus agalactiae were 42.3 mm, 35.2 mm and 41.5 mm respectively. The bacteriostasis effects of huanglianpu injection on these three pathogens were very sensitive. The resuLts of antibacterial experiment in vitro indicated that the antibacterial activity of huanglianpu injection on Staphylococcus aureus was stronger. Its MIC was 31.3 mg/mL,and MBC was 62.5 mg/mL, the antibacterial activity of huanglianpu injection on Escherichia coli and Streptococcus agalactiae were stronger, their MIC and MBC were 62.5 mg/mL. This provided the scientific evidence for filtering Chinese medicinal herbs to prevent and treat cow mastitis.By the methods of the mouse auricle inflammation, capillary vessel permeability and absorbent cotton induced granuLation, the anti-inflammatory effects of Huanglianpu injection on mastitis were investigated. By the methods of mouse immune organ index, macrophage enguLf chicken RBC, the abilities of the mouse phagocytic cell expurgating carbon, the immune functions of Huanglianpu injection on mastitis were studied. By the ameliorate Karber and half lethal dose (LD50) as evaluation acute toxicity intensity index, the injection acute toxicity was studied. The heat sources, excitant of this injection were also detected. The resuLts showed that mouse auricle inflammation swelling degree in high, middle dose groups of huanglianpu injection were 1.24 mg, 1.78 mg respectively, and compared with control group, the difference was significant (P<0.01). For OD values of capillary permeability, those in high, medium and low doses groups were 0.05, 0.14, 0.14, respectively and compared extremely significant difference (P <0.01), and compared with control group, the difference was significant (P<0.01). The mouse granuLation tissue weight in high, medium and low doses groups were 10.18 mg, 8.10 mg, 10.16 mg, respectively, and compared with control group, the difference was significant (P<0.01). The mouse thymus index, spleen index in the high, medium and low dose groups increased significantly (P<0.01) compared with control group. The abilities of macrophage in mouse abdominal cavity enguLfed RBC in high, medium dose groups, had significant difference (P<0.01) compared with controls group. In high-dosage group, the expurgation ability of mouse phagocytic cells on carbon particles had significant difference (P<0.01) compared with control group, but there were no significant difference in medium, low dose. In acute toxicity test, LD50 was 11222.77 mg/kg. The data showed huanglianpu injection had significant anti-inflammatory effects on acute and chronic inflammation treatment, couLd increase the mouse immune function, it couLd be used safely and reliable.Mastitis model was duplicated by infusing 104CFU/mL suspension of colon bacillus into one side of udder chamber of healthy rabbits. After modeling, the rabbit temperature rose, the heart rate and respiratory rate significantly increasd, the breast tissue presented the symptoms of the red, the swollen, the fever and the ache. After 3-7d treatment, the rabbit temperature, heart rate and respiratory rate started restoring in 3d treatment. These indices restored to normal levels after 7 days. There were no significant difference on RBC and HGB(P>0.05)before and after the normal levels. modeling. WBC, LYM and GRA changed significantly (P<0.01 ) with 3d treatment, but 7 days later they restored to the normal levels. The resuLts indicated that huanglianpu injection can treat rabitt mastitis.The studies showed huanglianpu injection couLd effectively strengthen the activities of SOD,GSH-Px,degrade concentration of free radical NO,MDA in sera and whey in clinical mastitis cows,to obtain the objective for treatment of cow mastitis.
