Traditional Chinese medicine has excellent therapeutic effect on the clinical inflammation of veterinary medicine.The research on the anti-inflammatory action and mechanism of traditional Chinese medicine are the hot topic.Gongyingsan is a classic prescription for the treatment of cow mastitis.However as a traditional Chinese medicine pulvis,it has defects,which are medication inconveniently,low availability,pesticide effect unruly,etc.And it has important scientific value and clinical significance that the anti-inflammatory effects and mechanism are studied for developing the newly dosage form of Gongyingsan powder,which can enhance the cure rate of livestock diseases,the modernization and application of traditional Chinese medicine in veterinary clinic.In this study,the classical methods were used to extract the effective components of each traditional Chinese medicine.Based on the analysis of ultraviolet absorption spectra,the compound injection of Gongyingsan was screened and prepared.According to the related requirements of Chinese veterinary medicine,the quality standard and safety of the injection was evaluated.The anti-inflammatory action and mechanism of the compound injection were studied by the mouse acute and chronic inflammatory model and levels of inflammatory factors in the serum of the tested mice.To evaluate the therapeutic effect of the compound injection on the recessive mastitis of dairy cows,the number of somatic cells in the milk was measured.Taraxacum mongolicum,Lonicera japonica,Forsythia suspensa,Folium Hibisci Mutabilis and Fritillary were used to prepare the compound injection.The p H,clarity,heat source,tannin,oxalate and protein of the injection were all in accordance with the quality standard of Chinese veterinary medicine.The results of the test showed that the compound injection of Gongyingsan could significantly inhibit the inflammatory models,which were the xylene-induced mice ear edema,acetic acid-induced capillary permeability increase and cotton-induced granuloma in mice,with the inhibition rates of 66.06%,47.97% and 25.92%.The compound injection of Gongyingsan could significantly increase the level of IL-10 in the serum of mice(P<0.01)and significantly reduce the levels of TNF-?,HMGB-1,PGE2 in the serum of mice(P < 0.01).The cure rate and effective rate of the compound injection of Gongyingsan were 50.00%,77.78% on the recessive mastitis of dairy cows,which had the outstanding clinical effect.In this study the compound injection of Gongyingsan was successfully prepared.Its anti-inflammatory action was clarified.It also preliminarily revealed its anti-inflammatory mechanism,which laid theoretical foundation for the clinical application of the compound injection of Gongyingsan.
