ZD Preparations Curative Effect On Mice Soft Tissue Injury Model Anti-inflammatory Analgesia And The Influence Of Caspase-3

Acute non-open soft tissue injured was applied by the compound traditional Chinese medicine ointment ZD preparation to explore its mechanism of action,pathological observation of acute non-open soft tissue injury in mice treated with ZD preparation;establishment of mouse ear swelling,rat toe Swelling,mouse peritoneal inflammation model,study the anti-inflammatory effect of ZD preparation;establish acute non-open soft tissue injury model in mice,measure the content of TXA2 and PGF1? in plasma at different time points,and induce platelet aggregation in vitro by turbidimetry to determine the function of blood stasis and phlegm of ZD preparations,to study the effect of ZD preparation on the expression of Caspase-3 by Caspase-3 activity detection and immunohistochemistry,in order to explored the biology of Caspase-3 in soft tissue injury effect by ZD preparation.Result:1.Histopathological observation the test drug group has a good ability to repaired damaged tissues,which can accelerate the repair of damaged muscle fibers,inflammatory cells dissipate and improve the degree of congestion.2.In the study of anti-inflammatory and swelling effects:(1)ZD preparation can significantly inhibit xylene-induced mouse ear swelling,egg white swelling caused by egg white,and reduce the degree of swelling in the initial stage of inflammation.(3)ZD preparation significantly inhibit the increase of peritoneal capillary permeability induced by acetic acid and reduce the increase of capillary permeability in the early stage of inflammation.3.In the study of the role of promoting blood circulation and removing phlegm:(1)ZD preparation can reduce ADP-induced platelet aggregation and regulate platelet function in vitro(2)ZD preparation can reduce the content of TXB2 in plasma of model rats,increase the content of PGF1? in plasma of model rats,and inhibit platelet aggregation.4.In the study of the biological effects of Caspase-3 in soft tissue injury under the intervention of ZD preparation,the following results were obtained:(1)Caspase-3 activity test showed that the difference between the high dose group of ZD preparation and the negative control group was extremely significant on the 3rd day,and the difference was not significant compared with the positive drug group,and the time was regular.(2)Imrmunohistochemistry showed that Caspase-3 was rarely expressed on healthy muscle fibers under the intervention of ZD preparation,and was expressed in a large amount in damaged muscle fibers and inflammatory cells.Conclusion:ZD preparation can accelerate the repair of acute non-open soft tissue injury,has good anti-inflammatory and swelling,promoting blood circulation and removing blood stasis,and can treat soft tissue damage by regulating the expression of Caspase-3.
