This thesis is one part of National Natural Science Foundation of China (Grant No.30970487) "Spruce-fir forest decline mechanism in low-lying land in Eastern Mountain Area of Heilongjiang Province under the global change" and National Natural Science Foundation of Heilongjiang province (Grant No. C200919) "Spruce-fir forest decline mechanism in low-lying land under the global change". The survey plots were selected for natural regeneration of Picea koraiensis in the Daxing'anling and Xiaoxing' anling arer of 12 Picea koraiensis provenances distribution area,At the same time,To reveal the genetic diversity of Picea koraiensis,144 samples which collected fresh spruce needles from 12 provenances of this species have been examined by using RAPD markers. Their genetic diversity was analyzed based on the population updated results in this study. The results can finally provide evidence of the population level and the molecular level for the recession mechanism of spruce.The main results are as follows:12 Primers are used to analyze the genetic diversity of 12 Provenances of Picea koraiensis using RAPD markers.83 loci are detected in total.there is higher genetic variation among the provenances of Picea koaiensisin DNA level. Proportion of the total polymorphic loci has been reached 98.81%.The Percent age of Polymorphic loci among different Provenanc es varies from64.29%to85.71%.The Percent age of Polymorphic loci of ShuangFeng Provenance is the highest,and that of YouHao Provenance is the lowest, also the same results got by Nei index and Shannon index.Shannon's Information Index and Nei's Gene Diversity Index are used to estimate genetic diversity for the provenances of Picea koraiensis.Nei (H) was 0.3632,Shannon index (â… ) was 0.5405. The genetic differentiations for both of inter-provenance and intra-provenance are 27.72% and 72.28%, genetic between provenances index Gst=0.2772,gene flow coefficient is 1.3040 Nm,espectively. It also shows that there is close relationship between different provenances,varying from 0.8046 to 0.9237.On the basis of cluster analysis of genetic distance,12 provenances have been divided into three groups,in which the first group includes Xinqing,Wuyiling and Hongxing provenances; the second is Wuying,Youhao,Meixi,Wumahe,Dailing,Nancha and Shuangfeng provenances; and the last group contains Mengkeshan and Talin provenances.Population regeneration data shows that:Mengkeshan and Talin in Daxinganling and Xinqing,Wuyiling and Hongxing provenances in Xiaoxinganling are progress populations,Wuying,Youhao,Meixi,Wumahe,Dailing,Nancha and Shuangfeng provenances in Xiaoxinganling are declining population. The genetic distance clustering results with natural regeneration results showed a greater consistency, It can further Provided evidence of Picea populations regeneration.in each Picea distribution area.The results may further provide a data base for the recession mechanism of spruce, and also provide a scientific basis for spruce genetic diversity conservation and ecological restoration.
