Objective: To study pharmacokinetics of florfenicol following a single i.m. administration to healthy goats and CCPP goats, and the effect of Chinese herb Morus alba L.chitosan on pharmacokinetics of florfenicol in healthy goats and CCPP goats.Methods: 24 healthy goats were allocated into 4 groups of 6 goats each at random. The goats of group A and B keeping healthy were intramuscularly injected with single dose of florfenicol at 20mg.kg-1 , the goats of group B were fed to Morus alba L.chitosan, immediately; After being infected with CCPP artificially , the goats of group C and D were intramuscularly injected with single dose of florfenicol at 20mg.kg-1 , the goats of group D were fed to Morus alba L.chitosan immediately. The plasma concentration of florfenicol was determined by HPLC. Pharmacokinetics parameters were calculated by 3P97 PK computer program.Results: Concentration-time data of all groups were best described by a one-compartment open model(W=1/C2), and the main parameters and calculated-equations as follows.respectively: Group A: t1/2Ka=0.46±0.29h, t1/2ke=9.64±4.78h, Tpeak =1.80±0.71h, Camx =4.16±1.23mg.L-1,AUC=60.34±17.25mg.h.L-1 , C=6.57(e0.11t-e2.56t ); Group B: t1/2Ka =0.32±0.17h, t1/2ke =9.11±3.84h, Tpeak =1.51±0.53h, Cmax =3.63±1.50mg.L-1 , AUC=49.79±15.25mg.h.L-1, C =4.51(e-0.09t-e-2.71t); Group C: t1/2Ka=0.41±0.25h, t1/2ke=7.41±3.66h, Tpeak=1.57±0.57h, Cmax=4.64±2.15mg.L-1, AUC=54.39±20.65mg.h.L-1, C =6.81(e-0.12t-e-2.47t; Group D: t1/2La=0.25±0.03h, t1/2ke =10.69±1.63h, Tpeak=1.40±0.18h, Cmax=4.09±1.42mg.L-1,AUC=67.42±17.81mg.h.L-1, C =4.60(e-0.007t-e-2.77tConclusions: (1)The absorption of florfenicol following i. m. administration to healthy goats was almost complete, distribution was rapid and wide, and elimination was slow. (2)When the goats were infected with CCPP ,the disease did not influence notably pharmacokinetics of florfenicol. (3)Morus alba L.chitosan had no significant influence to pharmacokinetics of florfenicol in healthy goats and CCPP goats. (4)In clinical veterinary medicine, Morus alba L.chitosan and florfenicol are used to treat the CCPP would be a practical and potential methods, florfenicol i.m., τ=24.2h, X0=21.2mg.kg-1, X0= 20.0mg.kg-1 , one time per day, with 3-4 times.
