Mastitis is the most expensive disease in dairy cattle flocks, and somatic cell count (SCC) is an indicator for mastitis especially subclinical mastitis indicating. It is necessary to establish an efficiency method to inspect mastitis. There are many methods of SCC, such as CMT (California Mastitis Test, CMT), WMT(Wisconsin Mastitis Test, WMT, somatic cell count with electronic instrument and DMSCC. Although CMT owns easy operation, it can only obtain a relative arrange of somatic cell without precise number. While WMT can obtain exact number but its operation is complex. somatic cell count with electronic instrument needs professional count equipment which is very expensive. In this study, we took count of somatic cells in milk samples from 39 cows in three cattle farms, three times every farm, by using of fluorescence dying. The result showed acridine dying is an accurate method for rapid detection of somatic cells in dairy cattle, and it also showed there is significant difference among cows of different breeding condition, management. Finally, according to our result, we pointed out some advices to improve the status of the dairy herd health.In this study we also cloned cDNAs encoding Bos taurus interferon -Tau (IFN-Ï„) from the genomic DNA of Holstein cow and expressed it by the form of DsbA fusion proteins in Escherichia coli {E. coli) using PET-39b vector.
