Study On The Protective Effect Of N-acetyl-cysteine And VitE On Lipid Peroxidation Damage Induced By Arsenic In Rat

The aim of the experiment was to study the lipid peroxidation damage of arsenic and the antagonistic action of N-acetyl-cysteine and VitE on the damage induced by arsenic in blood, liver and kidney of rats. 64 Kunming rats were divided randomly into 8 groups: control, arsenic exposed group, NAC and VitE groups (including low, medial and high doses respectively). Eight weeks later, all rats were killed to examine the changes of some biochemical indices (the antioxidases SOD, GSH-Px, CAT and the production of lipid peroxidation MDA in liver and kidney and SOD,δ-ALAD, Hb, MDA in blood). The results showed as fellows:Compared with those of control, the activity ofδ-ALAD, SOD in blood and antioxidases in liver and kidney of arsenic exposed group were significantly lower (P<0.01); and the content of MDA in blood, liver and kidney were significantly higher (P<0.01), but the Hb content didn't changed obviously (P>0.05).Compared with those of arsenic exposed group, the activity ofδ-ALAD and antioxidases increased significantly (P<0.05 or P<0.01), and the content of MDA decreased significantly (P<0.05 or P<0.01) in groups of low, medial dose NAC and high dose VitE.The results demonstrated that arsenic restrained the activity ofδ-ALAD which was the key enzyme in the synthesis of hemachrome, but didn't change the Hb content. NAC and VitE could improve the the activity ofδ-ALAD that restrained by arsenic. Arsenic decreased the antioxidative ability and increased the damage of lipid peroxidation. The antioxidant NAC and VitE could restrain free radicals production and lipid peroxidations induced by arsenic, increase the activity of antioxidases, therefore protect animal against the lipid peroxidation damage.
