Mastitis is one of the highest cost disease in milk cow breeding. Treatment mainly adopt antibacterial nipple perfusion and mamma block therapy presently. Mouth perfusion was taken as priority in Chinese herb therapy, whereas methods of mamma skin external treatment are seldom applied. There is no report of mamma external diadermic preparation futhermore.Research object in the study is the Chinese medicine therapy which has a long history of clinical use in our lab. Optimal principles of therapy and preparation are confirmed by using orthogonal design method.Taking berberine as testing object, cow skin as barrier in thin layer chromatography technique, investigated the optimal external diadermic accelerator. Results of uniform design optimization shows that compound diadermic accelerator is the optimal diadermic accelerator, which made up of azone.propylene glycol:field-mint=5%:10%:4%.The result of drug sensitivity test by 0~48h external transmission liquid reveals that 0 ~ 48h external transmission liquid has effects of bacteriostasis on common pathogenic bacteria, such as bacillus coli, staphylococcus aureus and streptococcus agalactiae,which causing clinical mastitis.The experiment not only lends a experimental support for the development of Chinese veterinary durg, but also provides a new route of administration for the treatment of clinical mastitis.
